Studies On Chemical Constituents Of Gankang Formula And Hepatoprotective And Jaundice-relieving Effects

Gankang Formula, a pulvis of Hainan island folk medicine complex, consists of three kinds of Chinese medicine such as Polygonum chinense L., Clerodendron cyrtophyllum Turcz and Herba hydrocotyles and has a good effect on treatment of icteric hepatitis patients. To our knowledge, the chemical constitutes and pharmacological activities of the folk medicine complex have never been reported previously. In order to take comprehensive utilization, it is very useful for studies on chemical constituents and pharmacological activities of the folk medicine complex.The air dried Polygonum chinsnse L., Clerodendron cyrtophyllum Turcz and Herba hydrocotyles were crushed, and then extracted by75%ethanol (in volume fraction). After evaporation under reduced pressure a dark ethanol extract was obtained. The extract was mixed with water, extracted by petroleum ether, chloroform and ethyl acetate, respectively. The chloroform soluble fraction was respectively subjected to column chromatography over macroporous resin, normal and reversed-phase silica gel, sephadex LH-20, preparation thin-layer chromatography and recrystallization repeatedly to obtain pure compounds. The chemical structures were established on the basis of extensive spectroscopic (IR,1D-NMR,2D-NMR, MS) data analysis and by the comparison with spectroscopic data reported in the literature. Then a test on TBIL and ALT of different fractions had been taken.(?)23compounds were isolated from chloroform fraction and identified as:β-sitosterol(1), palmitic acid(2),3,4-dimethoxyphenyl-ethyl-ether(3), diosgenin(4), betulinic acid(5), ursolic acid(6), stigmast-4-ene-3-one(7), crotafuran B(8),4-dimethylamino-benzaldehyde(9), genistein(10), tetrahydropalmatine(11), vanillin(12),2-butylated-4-hydroxyanisole(13), vanillic acid(14), emodin(15), leptosphaepin(16),4’-dihydrophaseic acid(17), gastrodin(18),4-caffeoyl quinic acid(19),arbutin(20),(Z)-resveratrol(3,5,4’-trihydroxystilbene)-3-O-β-glucoside(21), mannitol(22) and β-daucosterol(23). Besides compound (1)、(4)、(6)、(10)、(12)、(14)、(22)、(23), the others were isolated from Gankang Formula for the first time. And the75% ethanol extract fraction, chloroform soluble fraction showed strong activities onHepatoprotective and Jaundice-relieving effects (P﹤0.01). But the others showed weakactivities on Hepatoprotective and Jaundice-relieving effects (P﹥0.05).
