Objective: To investigate the expression and significance of CXCR4(ChemokineCXC motifreceptor4) and LKB1(STK11,serine Pthreonine protein kinase11) innon-small cell lung cancer and normal lung specimens taken from non-cancerousregions adjacent to lung cancer tissue, and their correlation with clinicopathologicalfactors as well as explore the role of them in the occurrence and development of nonsmall cell lung cancer,in order to provide a new theoretical basis for determining it’sinvasion/metastasisand prognosis of non-small cell lung cancer.Methods:1. the expression of CXCR4and LKB1in65cases of non-small cell lungcancer tissue (36squamous cell carcinomas and29adenocarcinomas) and22cases ofnormal lung specimens taken from non-cancerous regions adjacent to lung cancertissue were examined by EliVision immunohistochemistry. And analyse their relationswith age、gender、pathological types、pathological grade、stage and metastasis oflymph node.2. Statistical analysis was performed by spss16.0for windows. All testswas conducted at a significance level of a=0.05.Results:1. CXCR4expression was mainly localized in the membrane and cytoplasm, LKB1localized in the cytoplasm and nucleus, both of which indicate buffy or brown.2.(1) In NSCLC, the positive rate of CXCR4was significantly higher than thecases of adjacent normal tissue, the difference was statistically significant (P<0.05),(2) In NSCLC tissues, the expression or CXCR4is closely related toTNM stage and lymph node metastasis (P <0.05), but not to gender, age,histological type and histological grade (P>0.05).3.(1) In NSCLC, the positive rate of LKB1was significantly lower than the cases of adjacent normal tissue, the difference was statistically significant (P <0.05),(2) In NSCLC tissues, the expression or LKB1is closely related to histologicaltype、TNM stage and lymph node metastasis (P <0.05), but not to gender, age,and histological grade (P>0.05).4. A negative correlation is observed between the expression of CXCR4and that ofLKB1(rs=-0.262, p<0.05).Conlusion:1. The expression of CXCR4in NSCLC is abnormal, and the abnormal expressionrate was significantly higher than that in the normal lung specimens taken fromnon-cancerous regions. CXCR4may play an important role in the the occurrenceand development of NSCLC. And the overexpression rate was related to TNMstage and lymph node metastasis,which may become one of the indicators judgedmetastasis and prognosis of NSCLC.2. The expression ofLKB1in NSCLC is abnormal, and the abnormal expressionrate was significantly lower than that in the normal lung specimens taken fromnon-cancerous regions,which proves LKB1may participates in carcinogenesis ofNSCLC,especial of adenocarcinomas. The lower-expression of LKB1is related tohistological type、TNM stage and lymph node metastasis,which shows thatLKB1may be associated with infiltration、metastasis and prognosis of NSCLC.3. The negtive correlation between increased CXCR4expression and decreasedLKB1expression suggests that they might coparticipate in carcinogenesis andprogression of NSCLC through interacting with each other.4. The abnormal expression of CXCR4and LKB1are closely related to theoccurrence and development of NSCLC, and they may be used as referenceindexes for predicting the metastasis and the prognosis in NSCLC.
