Objective: By determining the expression of β-catenin and EZH2protein in renal clearcell carcinoma(RCCC) to investigate their significance and effect in RCCC occurrenceand development,and the correlating with RCCC cell clincopathological behaviors, andto provide a basis for clinical diagnosis,treatment and prognosis for RCCC.Methods: The expression of β-catenin and EZH2in50cases of RCCC and15cases inthe renal para-carcinoma tissues were determined with immunohistochemical S-Pmethod, as well as the correlation analysis of the expression of β-catenin,EZH2andtheir pathological grade, clinical stage, metastasis or not and other clincopathologicalbehaviors which combined with clinical materials.Results:1. β-catenin protein normally expressed in the membrane of renal tubular epithelialcells showed a yellow grid distribution, distribution, expression of the cytoplasm andthe nucleus less.β-catenin in renal cell carcinoma usually presents different: cellmembrane expression was decreased.In renal clear cell carcinoma of the positive rate ofcytoplasmic was78.0%(39/50). Cytoplasm of adjacent kidney tissue, the positiveexpression was33.3%(5/15),which was significantly lower than the rate ofcancer(p<0.05),that was to say, the expression in the tissues of renal cell carcinoma forβ-catenin was significantly higher than the renal para-carcinoma tissues;The positiverate for lymph node metastasis was95.5%(21/22),otherwise, the positive rate for nolymph node metastasis was64.3%(18/28), it showed that, the expression for β-cateninin the tissues of renal cell carcinoma was significant difference with lymph nodemetastasis(P<0.05); The positive rate for female was70.5%(13/17), The positive rate formale was78.8%(26/33), it meant, the expression for β-catenin in the tissues of renal cell carcinoma had no significant difference with sex(P>0.05);The positive rate for below50years was84.2%(16/19), The positive rate for above50years was74.2%(23/31), theexpression for β-catenin in the tissues of renal cell carcinoma had no significantdifference with age(P>0.05); β-catenin in renal cell carcinoma histological grade(Fuhrman grade) I Class, II-and III-level positive expression rate was64.3%(9/14),70.0%(14/20) and100.0%(16/16), with the pathological grade of the temperatureincreased, with statistical significance (P <0.05); β-catenin in renal clear cell carcinomaof the limitations of clinical staging of locally advanced renal cell carcinoma andmetastatic renal cell kidney cancer+positive rates were66.7%(18/27),91.3%(21/23),with the clinical stage of the temperature increased, with statistical significance (P<0.05).2. EZH2protein,which mainly expressed in renal clear cell carcinoma of the nucleus,the nucleus shows diffuse expression, more pale yellow and brown, its positiveexpression in the RCCC was74.0%(37/50), in adjacent kidney tissue positiveexpression rate was26.7%(4/15), significantly lower than cancer, that EZH2expression in renal cell carcinoma was significantly higher than the adjacent kidneytissue, the difference was significant (P <0.05); EZH2expression in the female grouprate of76.5%(13/17), The positive rate for male was72.7%(24/33), it meant, theexpression for EZH2in the tissues of renal cell carcinoma had no significant differencewith sex(P>0.05); The positive rate for below50years was67.7%(16/19), The positiverate for above50years was84.2%(21/31), the expression for EZH2in the tissues ofrenal cell carcinoma has no significant difference with age(P>0.05); EZH2in renal clearcell carcinoma histological grade (Fuhrman grade) well differentiated, moderatelydifferentiated and poorly differentiated, the positive expression rate was50.0%(7/14),80.0%(16/20) and87.5%(14/16), that is, the higher the tumor grade with EZH2inrenal clear cell carcinoma of the higher, with a significant difference (P <0.05);EZH2inrenal clear cell carcinoma limited to the board of the clinical stage and locally advancedrenal cell carcinoma metastatic renal cell kidney cancer+positive expression rate was63.0%(17/27),87.9%(20/23) not statistically significant (P=0.054);The positive ratefor lymph node metastasis was90.9%(20/22),otherwise, the positive rate for no metastasis was60.7%(17/28), it showed that, the expression for EZH2in the tissues ofrenal cell carcinoma was significant difference with lymph node metastasis(P<0.05).3.In50cases of RCCC, the positive expression of β-catenin was39cases, the positiveexpression of EZH2was37cases; There were7cases of RCCC that show positiveexpression for β-catenin protein and negative expression for EZH2, and5cases ofRCCC that show negative expression for β-catenin protein and positive expression forEZH2, and32cases of RCCC that show positive expression for β-catenin and EZH2,and6cases of RCCC that show negative expression for β-catenin and EZH2. Theexpression of β-catenin was positively correlated with the expression of EZH2inRCCC(rs=0.346, P<0.05).Conclusions:1.The expression of β-catenin and EZH2in Renal clear cell carcinoma was sigificantlyhigher than the adjacent renal tissue,suggesting thatβ-catenin and EZH2may played animportant role in RCCC occurrence and development.2.The expression of β-catenin and EZH2in RCCC was correlated with Pathologicalgrade and metastasis,and the expression of β-catenin was also correlated with clinicalstage.It suggested that expression of β-catenin and EZH2may associated with invasionand metastasis of RCCC.3.In Renal clear cell carcinoma group,expression of β-catenin had a positive significantcorrelation with expression of EZH2,however the relationship is not veryclose.,suggesting the both played a synergistic role in RCCC formation anddevelopment.β-catenin and EZH2may be used as an important endex to endicate theprognosis of Renal clear cell carcinoma.
