AIM To observe the effect of ω-3PUFAs on nuclear factor (NF)-κBexpression in the lung tissue of CLP induced acute lung injury rat model.Study theeffect of ω-3polyunsaturated fatty acids on NF-κB expression in sepsis inducedacute lung injury.METHODS Thirty experimental rats were randomly divided into three groups:healthy controls(A),sepsis group(B),intervention group(C).The sepsis acute lunginjury rat model of group B and C was established through cecal ligationperforation,and then,give group C nutritional support containsω-3PUFAs throughthe jugular enous catheter.The expression of NF-κB in the lung tissue of threegroups was detected respectively by immunohistochemistry and Westernblotting.Without cecal ligation and puncture.Group A do the same operationof group B.RESULTS In the pulmonary tissue of healthy controls rats,only a fewscattered NF-κB nuclear positive cells can be found;In group B,plenty of NF-κBnuclear positive cells are present in the airway mucous membrane,interstitiallung,alveolar space and endothelial cells(p<0.01);In ω-3PUFAs treatment group,the quantity of NF-κB nuclear positive cell is higher than that in the control group,but is decreased significantly than that in ALI group(P<0.01).Western blottingresults indicate that, the expression of NF-κB protein of the three groups isdifferent,ALI group and ω-3PUFAs treatment group have a higher expression thancontrol group, and at the same time,ALI group is higher than the ω-3PUFAstreatment group.CONCLUSION ω-3PUFAs can decrease the protein expression of NF-κB in the lung tissue of CLP induced acute lung injury rat model. ω-3PUFAs couldalleviate inflammation by inhibiting activation of NF-κB.
