With the increasing pressure of social competition and the expansion of the aging society, the memory-enhancing dietary supplements are entering people’s daily life. And the types of dietary food aiming at treating and preventing elderly neurodegenerative diseases are increasing. However, the phenomena of adding illicit medicines to dietary supplements still repeatedly happened, and the methods for selecting and detecting are relatively passive and lagged. The side effects of taking synthetic drugs immensely endangered people’s health (especially those elderly whose renal or liver function were diminished). Therefore, it is significant to establish an effective, rapid and real-time mean to screen the prohibited additives. In this paper, by summarizing the concept and effect of memory-enhancing dietary supplements, including the classification, the mechanism of action and typical drugs of nootropic, we concluded that high-performance liquid chromatography and mass spectrometry technology have more advantage in drug testing field by compared with the common detecting methods. Then using high performance liquid chromatography-Diode array detector (HPLC-DAD) and high performance liquid chromatography-mass spectrometry (HPLC-MS) method to analyze the potentially illicit medicines in memory-enhancing dietary supplements and compare the response of the target and characteristic fragment ions under different mass spectrometry conditions, and preliminarily establishing mass spectral library to monitor the nootropic. We hope that we can achieve an effective for screening and monitoring the illegally added drugs.The main contents are included as follows:1. The responses of six kinds of familiar nootropic drugs under different conditions of mass spectrometry are analyzed. The six kinds of drugs with similar Molar concentration use different mobile phases (methanol:water=1:1, and acetonitrile:water=1:1), and flowing inject into the different ion sources (ESI source, APCI source and DCBI source), then the corresponding spectra and characteristic fragment ions are acquired. By compared with the MS behavior of each material, we can analyze the main cause of the discrepancy of responses, then providing standards and abundant MS information for science.2. Establish a method of using HPLC-DAD to determine12kinds of potential nootropic drugs in the memory-enhancing dietary supplements. By optimizing the liquid chromatography separation conditions of12kinds of nootropic drugs, we can fast accurately and simultaneous determinate suspected additives in complex samples according to the retention and DAD spectra. The disposal method of sample is simple, and it has low detection limit, high reliability, high sensitivity and wide application. It also can provide a powerful basis for the quality supervision department.3. Develop a method of determination of11kinds of calcium antagonist drugs in the dietary supplements by high performance liquid chromatography-mass spectrometry. By optimizing the liquid chromatography separation and MS conditions of11kinds of drugs, the target has a better separation and mass spectrometry response. By the use of MS for qualitative and quantitative analysis, the result has a high accuracy, low limit and high sensitivity. By parsing the splitting rules of MS, a reference for adding these drugs or structure derivatives into the unknown samples is provided.4. The multi-stage mass spectrometry library of auxiliary improving memory drugs has been established. In order to establish an effective method to filter illicit drugs, it is necessary for our laboratory to establish a high accuracy and plentiful MS information data-base. The unknown sample can be known according to the spectra of these features (including one, two and multi-stage MS figure), then using collected information to match suited drugs searched in the case of the lack of reference substances or illegally added unknown drugs. It can achieve the purpose of initially filtering and confirming unknown composition within a short time.
