Oxidative stress represents an imbalance between the systemic manifestation of reactive oxygen species and a biological system’s ability to readily detoxify the reactive intermediates or to repair the resulting damage. Disturbances in the normal redox state of cells can cause toxic effects through the production of peroxides and free radicals that damage all components of the cell, including proteins, lipids, and DNA. Lots of studies have reported that many pathophysiologic progresses such as neuro-degenerative disease and aging are related to ROS. Recently studies on the mechanism of oxidative stress and the function of genes involved in it, the screening of antioxidative drugs have become a hot pot in oxidative stress filed.This study use the model organism C.elegans to carry out related workFistly, we developed a new oxidative stress model using HC10. Detecting the ROS level after HC1lO treatment and the protective function of antioxidant NAC.Furthermore, we detected the sensitivity of different mutants in HClO induced oxidative stress. The results show that HClO can be a new oxidative stress inducer in C.elegans and daf-2, egl-9, hif-1. vhl-1may respond in the oxidative stress pathway induced by HClO.Secondly, we constructed glb-13Is strain, the expression level of glb-13increased after PQ treatment and hNgb can protect C. elegans insdead of GLB-13. So our result shows that glb-l3is associated with reactive oxygen species (ROS) resistance evoked by paraquat stress as well as hNgb.In one word, our study develop a new oxidative stress model in C.elegans, rich the mechanism of oxidative stress study in C.elegans.
