Objectives:We detected the expression levels of Bcl-rambo, Rambo-beta, Xiap and Survivin mRNA in adult acute leukemia, to investigate the relationship between these genes’expression levels and the prognosis of acute leukemia, as well as the correlation between the expression level of Bcl-rambo, Rambo-beta, Xiap and Survivin mRNA, trying find out important molecular markers for evaluating the severity and prognosis of acute leukemia.Methods:Using Fluorescent quantitative RT-PCR method to detect the expression levels of Bcl-rambo, Rambo-beta, Xiap and Survivin mRNA in different risk group of adult acute leukemia patients (including low risk AML group, middle risk AML group, high risk AML group, standard risk ALL group and high risk ALL group), and in10healthy adults as control group.Results:1The expression level of Bcl-rambo mRNA in AL patientsIn AML patients, the expression level of Bcl-rambo mRNA in low risk AML group, middle risk AML group, high risk AML group was:0.7937±0.2542,1.1495±0.1970and1.4071±0.9292, respectively, which was statistically significant higher than the0.1942±0.1346in control group, P<0.05. The expression level of Bcl-rambo mRNA in middle risk AML group and high risk AML group was respectively higher than that in low risk AML group, P<0.05. But there was no significantly difference in Bcl-rambo mRNA expression level between middle risk AML group and high risk AML group, P>0.05.In ALL patients, the expression level of Bcl-rambo mRNA in standard risk ALL group and high risk ALL group was:1.3234±0.4985and 5.0910±2.3148, respectively, which was statistically significant higher than that in control group, P<0.05. The expression level of Bcl-rambo mRNA in high risk ALL group was statistically significant higher than that in standard risk ALL group, P<0.05.2The expression level of Rambo-beta mRNA in AL patientsIn AML patients, the expression levels of Rambo-beta mRNA in low risk AML group and high risk AML group were:1.7859±0.5014and1.8896±0.6975, respectively, which was statistically significant higher than the4.0766±0.9747in control group, P<0.05. But there was no significantly difference in Rambo-beta mRNA expression level between control group and middle risk AML group, P>0.05. And there was no significantly difference between low risk AML group and high risk AML group, P>0.05.In ALL patients, the expression level of Rambo-beta mRNA in standard risk ALL group and high risk ALL group was:8.2767±2.8793and9.6133±2.2289, respectively, which was statistically significant higher than that in control group, P<0.05. But there was no significantly difference in Rambo-beta mRNA between standard risk ALL group and high risk ALL group, P>0.05.3The expression level of Xiap mRNA in AL patientsIn AML patients, the expression level of Xiap mRNA in low risk AML group, middle risk AML group, high risk AML group was:0.6261±0.0899,0.6647±0.0870and0.8932±0.0737, respectively, which was statistically significant higher than the0.3135±0.0905in control group, P<0.05. The expression level of Xiap mRNA in high risk AML group was statistically significant higher than that in low risk AML group and middle risk AML group, P<0.05. But there was no significantly difference in Xiap mRNA expresson between low risk AML group and middle risk AML group, P>0.05.In ALL patients, the expression levels of Xiap mRNA in standard risk ALL group and high risk ALL group was:0.6248±0.0897、0.8239±0.1065, respectively, which was statistically significant higher than that in control group, P<0.05. The expression level of Xiap mRNA in high risk ALL group was statistically significant higher than that in standard risk ALL group, P<0.05.4The expression level of Survivin mRNA in AL patientsIn AML patients, the expression level of Survivin mRNA in low risk AML group, middle risk AML group, high risk AML group was:4.3413±1.5531,2.3889±1.0404and7.1496±2.1069, respectively, which was statistically significant higher than the0.4470±0.4173in control group, P<0.05. The expression level of Survivin mRNA in high risk AML group was statistically significant higher than that in low risk AML group and middle risk AML group, P<0.05. The expression level of Survivin mRNA in middle risk AML group was statistically significant higher than that in low risk AML group, P<0.05.In ALL patients, the expression level of Survivin mRNA in standard risk ALL group and high risk ALL group was:7.3163±1.9003and11.8942±3.4364, respectively, which was statistically significant higher than that in control group, P<0.05. The expression level of Survivin mRNA in high risk ALL group was statistically significant higher than that in standard risk ALL group, P<0.05.5The correlation between the expression level of Bcl-rambo, Rambo-beta, Xiap and Survivin mRNAIn all acute leukemia patients, the expression level of Bcl-rambo mRNA was positive correlated with that of Rambo-beta (r=0.480, p<0.05), that of Xiap (r=0.520, p<0.05) and that of Survivin (r=0.694, p<0.05); the expression level of Xiap mRNA was positive correlated with that of Survivin (r=0.649, p<0.05); but there was no linear correlation between the expression level of Rambo-beta mRNA and Xiap mRNA, and there was no linear correlation between the expression level of Rambo-beta and Survivin mRNA.Conclusions:1In AML patients, the expression level of Bcl-rambo, Xiap and Survivin mRNA in high risk AML group was statistically significant higher than that in control group and low risk AML group, which indicated that the expression levels of these three genes were closely related to the severity of AML. These three genes might be important molecular markers for evaluating the prognosis of AML2In ALL patients, the expression levels of Bcl-rambo, Xiap and Survivin mRNA in high risk ALL group were statistically significant higher than that in standard risk ALL group and control group, which implied that the expression levels of these three genes were closely related to the progression of ALL These three genes might be important molecular markers for evaluating the prognosis of ALL3No significant difference in the expression level of Rambo-beta mRNA was found in different risk AL groups, which prompted that the expression level of Rambo-beta mRNA in acute leukemia is non specific.4A highly positive correlation was found between the expression levels of Bcl-rambo mRNA and Xiap, Survivin mRNA. We surmise the reason for this phenomenon is due to the functionally related regulation. A highly positive correlation was obtained between the expression levels of Xiap mRNA and Survivin mRNA. We hypothesized that the reason for this phenomenon is that there is a synergistic effect between these anti-apoptosis genes.5The high expression of Bcl-rambo, Xiap and Survivin mRNA could be used as poor prognostic fators in acute leukemia.
