Objective: Thyroid is relatively active, its metabolism also increaseaccordingly, by in vivo hypothalamic-pituitary-thyroid axis and placenta-thyroid axis in the dual regulation and placental hormones, bloodconcentration of iodine, immunosuppressive effects in pregnancy, oftenincluding thyroid enlargement, cardiac output increased, peripheralvasodilation, thyroid hormone level and its immune changes of pregnancy, sothat the period of TT3, TT4were higher in normal reference value, and moreprone to hyperthyroidism in pregnancy.The increase of thyroid hormoneleading to a nerve, muscle stimulation, even consuming too much in patientswith hyperthyroidism in pregnancy. if the disease is severe,it can endanger theheart, causing heart failure. The research shows, maternal thyroid hormonelevel directly influences the growth of the fetus, thyroid hormone from theplacenta to the fetus in the entire pregnancy, the fetus itself due to thehypothalamus-pituitary-thyroid axis feedback function is not yet mature,causing thyroid internal environment disorder, make fetal organs developmentand function adversely affected. The low birth weight,and prematurity havehigh occurrence rate, and neonatal, perinatal mortality increases.But there isno story about while the heart is not affect offspring in different growth periodand how to impact at home and abroad. The study shows phosphorylation ofAkt (P-akt) and cardiac sarcoplasmic calcium pump (Serca), plays animportant role in cardiac hypertrophy and remodeling and may promotemyocardial apoptosis, possibly induce myocardial compensatory hypertrophyto heart failure conversion mechanism. myocardial diastolic time wassignificantly shorter, heart rate faster. Whereas the activity of cardiacendoplasmic reticulum Ca2+2ATP enzyme is in relation to thyroid hormone levels. While P-akt and Serca have expression in hyperthyroid heart diseaseearly stage,so they can be used as predictors of hyperthyroid heartdisease.Objective to research the effect on heart of offspring in the degrees ofmaternal hyperthyroidism,by observing the expression of P-akt and Serca inthe heart of offspring in different growth stages,the foundation was providedfor the prevention and therapy for heart diseases in future genetations.Methods: A total of60healthy SD female rats were randomly divided into3groups:control group(gavage for normal saline) mild hyperthyroidism(gavage for L-thyroxine50ug/100g) severe hyperthyroidism (gavage forL-thyroxine150ug/100g) then cause the rats pregancy. The newborn male ratson0days,21days and60days were respectively selected to research.Tomeasure the thyroid function from the heart,and slaughter, quickly take theheart, removal of intracardiac blood filter paper, dry, stored at-80degrees ofrefrigerator. Take heart tissue in4%paraformaldehyde fixed,paraffin-embedded.Histological sections (4m thick), and the section mountedon slides coated with protein glycerin, ordinary HE staining tissue tochanges.To determine the expressions of P-akt and Serca in Western Blotting.Results1The thyroid function examination of pregnant rats: The level of FT3and FT4of the mild hyperthyroidism group and the severe hyperthyroidismgroup was more significantly increased than normal control group (P<0.05).The level of FT3and FT4of severe hyperthyroidism group was increasedsignificantly contrasting the mild hyperthyroidism group (P<0.05). The levelof TSH of mild hyperthyroidism group and severe hyperthyroidism group wasdecreased significantly than normal control group (P<0.05).The level of TSHsevere hyperthyroidism group was decreased significantly contrasting the mildhyperthyroidism group (P<0.05).2The thyroid function examination of postnatal rats: The level of FT3and FT4in21days mild hyperthyroidism group and sever hyperthyroidismgroup was higher than normal control group (P<0.01), TSH was lower than normal control group (P<0.01). The level of FT3and FT4in21days severhyperthyroidism group was higher than mild hyperthyroidism group (P<0.01),TSH was lower than mild hyperthyroidism group (P<0.01). The level of FT3and FT4in60days mild hyperthyroidism group was higher than normalcontrol group, TSH was lower than normal control group but both were notstatistically significant (P>0.05), The level of FT3and FT4in60days severhyperthyroidism group was higher than normal control group (P<0.01), TSHwas lower than normal control group (P<0.01). The level of FT3and FT4in60days sever hyperthyroidism group was higher than mild hyperthyroidismgroup (P<0.05), TSH was lower than mild hyperthyroidism group (P<0.05).3HE staining, The cell shape arrangement were normal, myocardial fiberstructure clear, uniform coloring, nuclei were oval, nuclear size uniform, islocated in the central of myocardial fibers in the control group and the mildgroup; In the severe group,0day、21days of myocardial fibers becamethicker, disordered, visible muscle bundles to interrupt a phenomenon.Myocardial cells are visible inside the vacuolar degeneration. Part ofmyocardial cell rupture, degeneration and necrosis, found around theinflammatory cells infiltration and fibroblast proliferation, gap widened,obvious proliferation of fibrous connective tissues, myocardial cross-sectionalarea increases.However in60days The cell shape arrangement were normal,myocardial fiber structure clear, uniform coloring, nuclei were oval, nuclearsize uniform, is located in the central of myocardial fibers.4western result, The level of P-akt and Serca were noticely,especially on0days and the21days in the severe group,while no difference between controlgroup and mild group on0days、21days and60days.Conclusion1Thyroid hormone through the placenta into the fetal blood circulation, resulting in neonatal hyperthyroidism.2The degree of maternal hyperthyroidism is more serious, the offspringsof hyperthyroidism is more obvious.3The expression of P-akt and Serca have no difference between mildhyperthyroidism and contron group;4The expression of P-akt and Serca is significant in newborn rats andmice heart of severe hyperthyroidism in pregnancy, but its expression with agehas reversibility.
