| Objectives:(1) To explore the method of in vitro on-site determination about Plasmodiumfalciparum to current Naphthoquine/Azithromycin Naphthoquine〠Azithromycinanti-malarial drugs; to provide Naphthoquine/Azithromycinã€Naphthoquineã€Azithromycinmeasurement board with medicinal stability, repeatability, and validity for no less than3months.(2)Sensibility in Plasmodium falciparum is monitored in endemic area of Yunnan Province,providing the sensitivity of the baseline dates of Naphthoquine/Azithromycin antimalarial drugto Plasmodium falciparum, and providing technical support for the plexus surveillance ofsensitivity.(3) The effects of using Naphthoquine and Azithromycin and the combination the twodrugs are compared in endemic areas; and whether synergy exists.(4) The effects of two drugsAzithromycin and Naphthoquine are evaluated in treatment, expecting to open new areas for thedevelopment of anti-malarial drugs.(5) Training postgraduate research methods and scientificresearch capacity.Methods The method in vitro micro-test current was used. This method is based on in vitroculture method by Trager and Jensen and evolved in1978to further establish in vitro micro-testfor anti-malarial drugs sensitivity by Rieckmann. It is based on the different impact of differentconcentration of drugs on the parasite growth and development, under the principle of whichdrugs can completely inhibit Plasmodium falciparum growth and development by certainconcentrations. It can evaluate the inhibitory action of anti-malarial drugs on the formation ofschizont in vitro by microscopic examination.Results: The experiment was performed in Yinjiang County, Dehong Prefecture insouth-western Yunnan province.25patients of Plasmodium falciparum malaria were selected and19blood samples were tested. The success of plates is15, with the success rate of78.95%.Single-drug Naphthoquine IC50is3.14, Azithromycin IC50is31.46, and compound-drug Naphthoquine/Azithromycin IC50is0.09/0.07.The compound drugs’ half inhibitory concentration(IC50) was significantly lower than unilateral; the inhibition rate of single Unilateral and that of thecompound was increased as the drug concentration increased. The inhibition of the compounddrugs inhibiting the parasite was significantly higher than unilateral, and the amount of usage wasthe compound drugs significantly reduced the amount of drug than unilateral.Conclusions:(1)Self-restraint Naphthoquine/Azithromycin Naphthoquineã€Azithromycindrug-coated plates have stabilization, sensitiveness, good repeatability and user-friendly and canbe promoted for use in the site;(2) The sensitivity of the cases is not same for the Naphthoquine;(3) Drug dosage can significantly be reduced when the two drugs are combined and whethersynergy exists. |
