Objective:To construct a model of acute graft-versus-host disease murine, aiming toclarify the mechnaism of CD45-of Nestin+stem cells fucntion in support of hematopoieticand mediated by HSCs homing. Methods: Construction aGVHD model, detectedchemokine levels of CXCL-12, SCF,IL-7and VCAM-1, detect difference expression ofCXCL-12, SCF, IL-7and VCAM-1in CD45-of Nestin+stem cells. Results: The aGVHDmodel was constructed successfully. Significant difference is existed between aGVHDmodel and BMT group from the level of CXCL-12, SCF, IL-7and VCAM-1, and Thelevel on gene and protein of these factors at different time points. The day when tookGFP-cell infusion into BMT and aGVHD group on beginning,+3days and+7days aftertransplantation, The bone marrow cell homing in BMT group was significantly higher thanthe GVHD group. There was no significant difference in two groups on the level of theCXCR4expression from HSC cells in the bone marrow of mice at different time pointsafter transplantation. Conclusion: The aGVHD environment lead to reduce bone marrowCD45-Nestin+cell count, the level of IL-7,VCAM-1, CXCL-12factors are in lowexpression state from genetic subject, CXCL-12/CXCR4mediated by HSCs homing beinduced dysfunction in aGVHD enviroment.Hence,aGVHD disease effect on the implationof HSC and hemopoiesis differention which change bone marrow Niche by impacting onCD45-Nestin+cells quantity and fuction.
