Objective: To observe the effect on mRNA expression of MAP-2and NF-L in ischemic penumbra of cerebral ischemia reperfusion rats treated by acupuncturing DU meridian, and to explore the neuroprotective mechanisms of acupuncturing DU meridian in ischemia reperfusion tissue.Methods:Rats were randomly divided into5groups:normal group, sham operation group, model group, DU meridian acupoints acupuncture group and Yangming meridian acupoints acupuncture group. Took the focal cerebral ischemia rats as rats model.3hours after finishing model-making, used the method of acupuncturing DU meridian acupoints which compared with acupuncturing Yangming meridian acupoints as means of intervention on rats. Then the changes of neurobehavioral scores were observed. Took the brain after24hours, and detected the changes of the mRNA expression of MAP-2and NF-L in ischemic penumbra by Real-time PCR method.Results:(1)Neurological score:Both the DU meridian acupoints acupuncture and the Yangming meridian acupuncture can significantly improve the neurological score before and after treatment compared with the model group(P<0.05), and the DU meridian acupoints acupuncture group has the better effect than the Yangming meridian acupuncture group(P<0.05).(2) mRNA expression of MAP-2and NF-L:MAP-2and NF-L in the brain tissue of the normal rats and the sham operation rats are on the level expression; and they are all significantly lower expressed in the model group than in the normal group(P<0.01). In the DU meridian acupoints acupuncture group and the Yangming meridian acupuncture group, the two mRNA expressions are higher than the model group(P<0.01), but lower than the sham operation group and the normal group, and the DU meridian acupoints acupuncture group has a more significant effect.(P<0.05).Conclusion:Acupuncturing DU meridian acupoints can significantly improve the neurological damage after ischemia-reperfusion injury of rats, and has more significantly effect on raising the mRNA expression of MAP-2and NF-L than acupuncturing the Yangming meridian. This effect could promote dendrites and axons to sprout and regenerate in brain after cerebral ischemia reperfusion, improve the plasticity of neurons and neurological status, finally protect the injured cerebral nerves.
