Objective:To prove the validity of the "the color to color" therapy for pigmentary diseases and provide theoretical basis for this therapy through respectively adopting B16melanoma cells culture and in vitro tyrosinase experiments as well as respectively observing the activation of20kinds of Chinese herb (a group white herb and a group of black herb) on tyrosinase activity. Methods:The laboratory cultivates the mice B16melanoma cells bought from Xiang Ya center cell laboratory, with blank group as the control group, confecting the20kinds of Chinese herbs (black group and white group) into three concentrations of drug culture medium, acts on the cells after a certain amount of time according to the different methods and then uses MTT method to determine the melanoma cell proliferation, NaOH cracking method to determine the melanoma cells melanin content and dopamine oxidation method to determine the tyrosinase activity. After that, the laboratory conducts in vitro tyrosinase experiment, making the3concentrations of the previous20kinds of Chinese herb act on the mushroom tyrosinase, determines the activate rate of the tyrosinase activity by way of adding intervention of different functions. Results:As a whole, among the20kinds of herbs, the black Chinese herbs have a much better activation function than the white ones on cell proliferation, melanin synthesis, tyrosinase activity as well as in vitro tyrosinase experiment. And it can be seen that part of the Chinese herbs have a dose-response relationship with the activation function of mice B16melanoma cells tyrosinase activity. Conclusion:Dark color Chinese herbs ((black, purple, red, purple red) have a more apparent effect on tyrosinase activity; while the light color herbs have a more apparent prohibiting effect. This shows that the color of the herbs have certain relevance with the metabolism regulation of pigment. Clinically, Chinese herb therapy is relatively safe and nontoxic; therefore dark color Chinese herbs can be used to treat the pigmentary loss diseases.
