Drug abuse is defied as a long-term use of psychoactive substances or enhanced medicine in a non-therapeutic or non-medical role. They can bring central nervous system with excitement or depression effect, resulting in a variety of hallucinations and making people’s thinking, mood or behavior out of control.Abuse of drugs and anti-mental disorders is not only harmful to human health, damage the brain and central nervous system, but also plays an important part in criminal cases. With the ability in controlling the human brain and central nervous system, drug abuse is used by the criminal to do anesthesia robbery, rape, fraud and other criminal activities, that do serious harm to the social and public safety. Drug abuse has gradually become a global social problem, along with economic development and the trend of economic globalization, the standards and requirements for physical and chemical inspection personnel for China’s public security departments inspection of drug abuse have risen, although the immune board and other rapid detection methods has a good prospect for a few criminal cases, but its versatility, as well as the lack of full sample information determine that it can not become the primary means of inspection and testing for drug abuse. Taking shorter metabolic half-life and other characteristics into consideration, the past detection methods to test the prototype drugs gradually withdrew from the stage of history, replaced by the detection of metabolism of the drug abuse which means deep-seated detection. Studying the metabolism of the drug abuse in vivo by suggesting the unidentified metabolites and drug metabolic pathways is of great significance in science prospect, and provides a strong basis for the fight against illegal and criminal activities.In this thesis, by using the biological model of rats and advanced analytical techniques as SPE, SPME, GC-MS, LC-MS, TOF-MS, the metabolites and metabolism of abuse drugs as chlorpromazine, zolpidem, amitriptyline, chlordiazepoxide and diazepam were studied. By comparation with blank urine sample、the standard and other methods, the in vivo metabolites and metabolic pathways in rats of the drugs were detected and resolved. The conclusion can be applied to the public security departments when doing physical and chemical inspection procedures for samples, providing a theoretical basis for the related criminal case.
