Objective To investigate the expression of IL-6and IL-10levels of subtypes lymphomaand the clinical significance in tissue and plasma. And we studied plasma levels of IL-10and single nucleotide polymorphisms in the IL-10(rs1800896) gene promoter in patientsof Han and Uygur with lymphoma to address potential genotype phenotype correlations.Methods Immunohistochemical technique and enzyme-linked immunosorbent assay(ELISA) were used to assess the expression of IL-10and IL-6in97cases paraffin tissueand132cases plasma samples of subtypes lymphoma,respectively. The SNaPshot singlenucleotide polymorphism (SNP) genotyping method was used to analyze the genotypes ofIL-10(rs1800896).Results Expression of IL-6and IL-10was high and there is no significant differencebetween subtypes lymphoma (P<0.05). Tumor samples were found positive IL-6andIL-10expression in macrophages and vascaular endotheial cells by immunohistochemistryrespectively. Expression of IL-6and IL-10in plasma of lymphoma was higher thancontrol, IL-6and IL-10in these lymphoma were significant difference (P<0.05). IL-6and IL-10plasma levels increase with enhanced expression in tumor cells (P<0.05), Theexpressions of IL-6and IL-10detected by immunohistochemistry were found correlated(ï½'=0.394,P<0.05). The plasm levels of IL-6in T-NHL were significant higher thanB-NHL (P<0.05), and other groups in different subtypes lymphoma were no significantlydifference(P>0.05). There existed a significant difference between the Kazak and Han,Uygur cases in levels in plasm IL-6(P<0.05). The expression of IL-6in the groups of Bsymptoms, high risk and higher levels of plasmβ2-MG, LDH, CRP were significantlyhigher than groups of no B symptoms, low risk and lower levels of plasmβ2-MG, LDH,CRP (P<0.05). The groups of Ann Arbor stage â…¢-â…£, B symptoms, and high expressionplasmβ2-MG and CRP were significantly higher IL-10levels than groups of Ann Arbor stage â… -â…¡, no B symptoms, and low expression plasmβ2-MG and CRP (P<0.05). Theexpression of plasm IL-6and IL-10were lower in CR patients than PR, DP and notreament groups(P<0.05). Expression of Plasm IL-10were positively correlated withIL-6and β2-MG (P<0.05). Frequency of the single nucleotide polymorphisms in theIL-10gene promoter have no significance in patients with lymphoma of Han and Uygurcompared with controls(P>0.05); The frequency of position rs1800896AG of the IL-10promoter was found to be significantly higher in patients of Uygur with lymphomacompared with controls(P<0.05). There was no significance in the single nucleotidepolymorphisms rs1800896in the IL-10gene of different lymphoma substype (Pï¹¥0.05).IL-10rs1800896AG genotypes had a significantly higher probability of freedom in higherIL-10plasma levels group.(P>0.05).Conclusion IL-6and IL-10had been found various degree expression in tissue andplasma of lymphoma. IL-6and IL-10in plasma of lymphoma were higher than control.These results show that IL-6and IL-10may cooperate in vivo to lymphoma cellproliferation and infiltrate. IL-6and IL-10may serve as useful indicators for diagnosis oflymphoma. Levels of plasm IL-6and IL-10concered with tumor load. The plasma levelsof IL-6and IL-10may be served as means to observe the lymphoma patients, tools topredict the response to therapy and prognosis. This analysis indicates that the geneticbackground can modulate plasma levels of IL-10.
