Objective:To evaluate the relationship between expression of Toll-like receptor4,7and9in Xinjiang Kazak and Han esophageal squamous cell carcinoma(ESCC)andwith local invasion and lymphnode metastasis. Methods:The mRNA of TLR4, TLR7andTLR9in40cases of ESCC and its adjacent tissues were detected with real-timefluorogentic quantitative PCR(qRT-PCR), and their protein detected with IHC methodfrom90cases of ESCC and its adjacent tissues. Results:The quantification of TLR7andTLR9mRNA in ECCC tissues were(0.72±0.59and0.07±0.06)respectively, which weresignificantly higher than in adjacent tissues (0.37±0.76and0.01±0.05) respectively (P<0.05),while no significant with TLR4mRNA in ESCC and its adjacent tissues (P>0.05).The quantification of TLR4,7,9mRNA in Han nationality ESCC was significantlyhigher than expression TLR4,7,9mRNA in Kazak ESCC (P<0.05).The proteinexpression of TLR4,TLR7and TLR9in ESCC were (66/90(73.2%),68/90(77.6%)å'Œ67/90(75.4%) respectively, which were significantly higher than that in adjacent tissues(18/90(20%),17/90(18.9%)and30/90(33.3%), P<0.05).There was a closerelationship between the protein expression of TLR4and lymph node metastasis (P<0.05).TLR7over expressed in ESCC was remarkably associated with tumor invasion (P<0.05). The protein expression of TLR9in Han nationality ESCC was significantly higherthan the protein expression of TLR9in Kazak ESCC (P<0.05).TLR4,TLR7and TLR9over expressed in esophageal cancer was remarkably related with tumor differentiationdegree (P<0.05).The higher protein expression of TLR4in ESCC interstitial cells wascorrelated with invasion and lymph node metastasis. The higher expression of TLR9infibroblast cells of ESCC interstitial was remarkably related with lower of invasion and lymph node metastasis (P<0.005).Conclusions: Our findings indicated that the higherexpression of TLR4, TLR7and TLR9in ESCC were significantly associated with ESCCcarcinogenesis, invasion and metastasis. These differences between ethnic groups mayreflect the different genetic background and the tumor pathogenesis.
