Cardamine Urbaniana0. E. Schlz blongs to Cardamine genus, which is perennial herb."Caiziqi" and "Banbiancai" are known as another name for this herb. It is a common traditional Chinese medicinal. Rhizome can be used as the medicinal part. It has the traditional effects of relieving cough and reducing sputum, blood circulation, antidiarrheal. It is used for the treatment of pertussis,chronic bronchitis, injuries from falls and so on.As the edible wild Chinese traditional medicine, it is characterized by highly economic values and wide application. But Cardamine Urbaniana0. E. Schlz has not been adopted by the Chinese pharmacopoeia and it’s Chemical constituent and pharmacological effect have not been reported now. It is only carried on related research on pharmacognosy and mineral element.In this thesis, L9(34) orthogonal experimental and variance analysis are applied to study the best extraction process of volatile oil in Cardamine Urbaniana0. E. Schlz, It considers the investigate factors of granularity of burdock dusts, water ratio, immersion time and extraction time. The best extraction process of volatile oil is that herbs by crushed through65mesh fineness,12times amount water, immerse6hours and sequential extract10hours, this volatile oil extraction ratio is highest.The study of extraction technology of volatile constituents in Cardamine Urbaniana0. E. Schlz provide scientific basis for the development and application.Volatile constituents in Cardamine Urbaniana0. E. Schlz is adopted by SD/GC-MS combination technology and HS-SPME/GC-MS co-mbination technology to compare and analyze, two kinds of metho-ds are identified75compounds, using the SD/GC-MS combination technology of volatile oil dentified35compounds, and the use of HS-SPME/GC-MS combination techniques are identified48compo-nents, which by the two methods together to identify the volati-le components of8kinds. The results show that,volatile consti-tuents in Cardamine Urbaniana0. E. Schlz contains a variety of aldehydes, ketones, sesquiterpenes, alkanes, fatty acids, isothiocy-anate ester, aromatic compounds and their derivatives and other chemical constituents. In volatile constituents from Cardamine Urbaniana O. E. Schlz, the content of4-isothiocyanato-l-Butene is hightest.Secondly, higher content of composition are4-Formyl-1,3(2H)-dihydroimidazole-2-thione,2,6,10-trimethyl-Penta-decan-e,5-propyl-Tridecane,4-Methylpentyl isothiocyanate, n-Hexa-deca-noic acid and so on.Chemical constituents of Cardamine Urbaniana0. E. Schlz are systematically investigated by silica gel chromatography, Sephadex LH-20chromatography,preparation of HPLC and recrystall ization. Three compounds are isolated from ethyl acetate extraction. The chemical structure is in the process of identification now.The TLC method is used for qualitative identification of Cardamine Urbaniana0. E. Schlz. Through the study of thin-layer conditions including extraction method,extraction solvent and developing agent, finally the program is set with silica gel G plate, chloroform-ethyl acetate-methanol-water (15:40:22:10) placed10℃below to clarify the lower solution as developing agent,spraying10%sulfuric acid in ethanol, heated to a clear spot color at105℃, respectively, set daylight and UV light (365nm) to view. The product chromatography, and the control drug in chromatographic corresponding place, respectively show the same color of the spot or fluorescent spots. The identification method is simple, convenient, quick, strong reproducibility and the results are reliable.The content of total saponins in Cardamine Urbaniana O. E. Schlz is detected by the UV-visible spectrophotometry. The method to prepare solution of product is determined after search of its extraction method,solvent multiples, the number of extraction,and extraction time. With oleanolic acid as reference substance,the content of total saponins is detected by the UV-visible spectrophotometry at547nm wavelength, meanwhile corresponding methodology investigation is carried out. By measuring the five batches of samples, tentatively set product of total saponins should not be less than1.81%.According to the pharmacopoeia of the People’s Republic of2010version of the measurement method of the appendix,respectively examine the volatile oil content, moisture, total ash and acid insoluble ash of Cardamine Urbaniana O. E. Schlz. Thus, the method is accurate, reliable, reproducible,which can preliminary establish a quality evaluation system of Cardamine Urbaniana O. E. Schlz.Chemical constituents and quality standard of Cardamine Urbaniana O. E. Schlz are systematically investigated.It is identified75volatile compounds. On the basis of above experimental results,quality specification draft and additional remarks detailed in draft was worked out.It provides the scientific bases for the quality of Cardamine Urbaniana0. E. Schlz, and it establishes a certain foundation to deepen its study. All these achievements are of great significance for rational development and utilization of Cardamine Urbaniana0. E. Schlz.
