A(?)m:Synthesize ethyl2-(4-iodobenzyloxy) acetate derivatives, and discuss these compounds’effects of learning and memory facilitation in mice. Methods:1-iodo-4-methylbenzene (3) was prepared by p-toluidine through diazonium salt method.1-(bromomethyl)-4-iodobenzene(4)was obtained by1-iodo-4-methylbenzene (3) through Whol-ziegler reaction with NBS. The compound(4) reacted with sodium2-ethoxy-2-oxo-ethanolate to give ethyl2-(4-iodobenzyloxy) acetate(5). The compounds8a-8t were obtained by the reaction of esterification with organic tin catalytic. on compound (5) with a series of substituted ethanols. The effects of compounds on learning and n emory in mice were tested by water maze test. Results:We designed and synthesized twenty new compounds, and the structures of the compounds were confirmed by IR.1H-NMR and MS spectrum. The pharmacological results showed that ethyl2-(4-iodobenzyloxy) acetate derivatives have strong learning and memory facilitation in mice.
