Objective:20(R)-ginseng saponins Rg3induces A549cells of human lung adenocarcinoma apoptosis. However, the precise mechanism and role are unclear. In this study,we will investigate them to provide further basic theory and experiment evidence of chemotherapy of non-small lung cancer treated by20(R)-ginseng saponins Rg3,facilitate the further development and clinical application of it.Methods:1. Median inhibitory concentration of the drug is analyzed by MTT,as well as the distinction of the proliferation of human lung adenocarcinoma A549cells with variance levels G-Rg3administration between24、72、120hour.2. The effect of different concentration G-Rg3treats human lung adenocarcinoma A549cells between72h and120h to cell cycle and apoptosis analyzed by flow cytometry.3. Western blotting examines the related-protein of human lung adenocarcinoma A549cells treated with G-Rg3expression.Results:1. Median inhibitory concentration of human lung adenocarcinoma A549cells treated with G-Rg3is740umol/ml;the inhibition ration has no significant compares to control group in human lung adenocarcinoma A549cells with variance levels G-Rg3administration between24h、72、120h.(p>0.05)2. Five concentration G-Rg3treat human lung adenocarcinoma A549cells after72h and120h,the cell ration of different cell cycle has no statistical significance compared with control group (p>0.05), meanwhile human lung adenocarcinoma A549cells treated with G-Rg3apoptosis ration have significant difference and exhibit time-dependence compared to control group by flow cytometry analyzed.3. Western blotting indicates:diversity time/concentration G-Rg3contribute human lung adenocarcinoma A549cells leading to activation of caspase-3/caspase-8connection, up-regulation Bax and Bid Cyto-c、down-regulation Bcl-2expression, presenting time-concentration independence.Result:1. G-Rg3induces human lung adenocarcinoma A549cells apoptosis, increasing the concentration and extending time of G-Rg3,the role of promoting apoptosis enhanced.2. G-Rg3mediates human lung adenocarcinoma A549cells apoptosis via up-regulation Bax、down-regulation Bcl-2protein expression, up-regulation Bid、 Cyto-c protein expression.3.The caspase-3/caspase-8connection is also the critical pathway which G-Rg3induces human lung adenocarcinoma A549cells apoptosis.
