Objective Probe the influence of collagen type Ⅱ (C Ⅱ) dose to modeling success rate, and detect the situation of distribution and expression of HMGB1in the normal control group and experimental group SD rats’synovia.Methods SD rats’back and base of the tail was injected intradermally the mixed emulsion maded of Bovine CII with incomplete Freund’s adjuvant (IFA) to induce RA animal models:collagen-induced arthritis (CIA) model.48male SD rats numbered were divided into four groups randomly, to probe the influence of0μg、200μg、300μ、400μg4C II dose level to modeling success rate. Every week, observate the situation:the appearance of swelling、weight and changing of the foot, to detect the arthritis index (AI). Every two weeks, using enzyme-linked immunosorbent assay (ELISA), to detect the serum concentration of anti-CII antibody and TNF-alpha, until rats were sacrificed on42th day after the initial immunization, with X-ray radiography and H-E staining of the pathology, to check the situation of joint disease and bone destruction. Select the SD rats in model group, to detect the situation of distribution and expression of HMGB1in synovia, using immunohistochemical staining method, and compare with the normal group.Result The sick SD rats were visible with weight slow growth or even decline, paws pad thickening, ankle swelling, and varying amounts of plantar and toe joint swelling, in addition, difficulty in mobility. In the acute disease phase, activities of ill feet reduced significantly, the average arthritis index (AI) reach5.15±1.82, the serum concentrations of anti-CII antibody and TNF-alpha increased more high than the control group, there was statistically significant,P<0.05. On the42th day after the initial immunization, X-ray and morphological changes were the performance of chronic synovitis, showed the bone with obvious invasion of the cloud. By statistical analysis, we can learned that modeling success rate increased with the CII dose, there was statistically significant, P<0.05, the modeling success rate of400μg group attained 100%.The process of modeling, All SD rats did not occur deaths in variety of CII dose. Immunohistochemical staining showed that the synovial HMGB1of normal control rats distributed mainly in the nucleus, but the CIA rat in the nucleus and the cytoplasm, and the expression increased, comparing with normal control group.Conclusion1.Bovine type II collagen can induce the SD rats’ animal model of rheumatoid arthritis, and increase the dose of bovine type Ⅱ collagen can improve the modeling success rate,between200μg to400μg;2.In the experimental group, the SD rats’synovial HMGB1distributed widely and expressed highly,suggesting that the HMGB1maybe have an important regulatory role in rheumatoid arthritis.
