Metabolomics is the most recent technology in the world of "Omics" joining genomics, transcriptomics and proteomics, in an area of science trying to evaluate tissues and biological fluids for changes in endogenous metabolite levels that result from disease or therapeutic treatment.The focus of this paper is how to effectively regulate the hyperglycaemia of the Type2diabetes mellitus. Two main research areas are as follows:(1) metabolomics was applied to the research of dynamic urine metabolic profiling and biomarkers of type2diabetic KK-ay mice treated with repaglinide based on GC-MS;(2) A molecularly imprinted solid-phase extraction method was developed to separate the resveratrol which could be used to improve the symptoms of the Type2diabetes mellitus from the Polygonum cuspidatum Sieb. et Zucc.The contents in this paper mainly includes the following parts:(1)GC-MS technique coupled with conventional derivatization protocols were used to analyze the metabolites profiling of urea from twenty diabetic KK-ay mice and five C57BL/6J mice. In the TICs,41peaks were considered to be endogenous metabolites. As for the quantification, peak area was employed. To normalize the urinary data, the variables were expressed as the ratio of peak area of the corresponding metabolites to that of the internal standard on the same chromatogram. The endogenous metabolites of mice urine were identified by the standard components and the NIST library. The difference of the endogenous metabolites which belonged to different mice models were found out by the quantitive and the qualitative work.(2)Investigated the therapeutic effect of repaglinide to the type2diabetes mellitus by analysising the5important endogenous metabolites. The results demonstrated that repaglinide not only regulate the sugars and polyalcohol but also the organic acid in the organism.(3)Using trans-resveratrol as the template molecular,4-vinylpyridine(4-VP) as the functional monomer, divinylbenzene(DVB) as the cross-linker, a molecular imprinted polymer with excellent preferential adsorption for resveratrol was prepared by bulk polymerization molecular imprinted technique for the first time. The particle size distribution, adsorption ability and selectivity of the molecular imprinted polymer was investigated. The results showed that, the MIP with regular shape and good dispersity; two kinds of binding sites were produced in the polymer matrix and their dissociation constants were calculated to be1.09×10-3mol/L,2.17×10-3mol/L, respectively. Excellent selectivity to resveratrol was exhibited on the MIP. The solid phase extraction experiment showed that the MIP was suitable for separating resveratrol from the Polygonum cuspidatum Sieb. et Zucc.Extracts.
