OBJECTIVE:To observe the neuroprotective effect of Rosuvastatin and its influence to Hypoxia inducible factor-1alpha (HIF-1) and Vascular endothelial growth factor (VEGF) in the model of experimental intracerebral hemorrhage (ICH), and to explore the possible mechanism that how the Rosuvastatin to protect nerve tissue。METHODES:Eighty Spragne Dawley (SD) rats were randomly selected and divided into2groups:ICH model group (A group), ICH rosuvastatin therapy (lOmg/kg)(Bgroup). To utilize stereotaxic apparatus and infuse fresh quantited autologous blood into the right basal ganglia to build up rat experimental ICH model; Treatment groups were administered with rosuvastatin(10mg/kg/d) for seven days. As comparing research, Shamed-operation group and ICH groups were administered with phasphate-buffered saline for seven days.After injecting,neural function defect of rats were assessment by measurement of Garcia’s score method.Then the SD rats were sacrificed, expression of HIF-1alpha and VEGF around hematoma in Rats’brain were analyzed by enzyme linked immunohistochemistry (IP). At24h,72h and7d, using dry and wet method for determination of the water content of brain RESULTS:1.Nerve function defect after cerebral hemorrhage, the symptoms reached the peak at48h.Neural function score of ICH group was lower than rosuvastatin intervention group, difference were statistically significant (P<0.05).2. The water content of brain was increased after intracerebral hemorrhage, each time point of rosuvastatin intervention group (group C) and cerebral hemorrhage model group (group B) compared to all have statistical difference (P<0.05).3. There were lot of HIF-lalpha and VEGF expressed after Intracerebral hemorrhage。 Each time point compared with the cerebral hemorrhage model group, expression of HIF-1alpha and VEGF was decreased in rosuvastatin intervention group, difference were statistically significant (P<0.05).CONCLUSIONS:1. Rosuvastatin can reduce the neural function defect of experimental intracerebral hemorrhage rat.2. Reduce the acute brain edema, ease the HIF-1alpha and VEGF expression level after intracerebral hemorrhage, may be the mechanisms of rosuvastatin to exert neuroprotective effects in inintracerebral hemorrhage.
