Objective:Combining EMT and CSCs theories, this study preliminarily researched on the impact of EMT on the biomarker of cell sternness and explored its significance.Method:1. Dynamic observations on the morphological changes of cell line ACC-M induced by TGF-β1:This study used TGF-β1to induce EMT in the SACC cell line ACC-M and then observe its morphological changes;2. Dynamic observations on the expression of biomarkers related to epithelial-mesenchymal transition and cancer stem cell in cell line ACC-M induced by TGF-β1:We employed flow cytometry (FCM) techniques to investigate the expression of EMT-related biomarkers E-cad, N-cad, vimentin and cell sternness-related biomarkers Oct-4in the TGF-β1-induced ACC-M cells; data was analyzed with SPSS11.0by using General Linear Model.Results:1. In vitro cell culture reveals that TGF-β1induced ACC-M cells undergo morphological changes from tightly arranged polygon to scattered spindle-like form;2. The expression of E-cad of both case and control groups does not satisfy sphericity test. After correction, the difference between the two groups is statistically significant (P<0.05), as well as that among different time points (P<0.001). There is interaction between time and grouping, meaning the longer the time of cell culture is the more apparent effect the TGF-β1would cause (P<0.05); The expression of N-cad of both case and control groups satisfies sphericity test. The difference between the two groups is statistically significant (P <0.05), as well as that among different time points (P<0.001). There is interaction between time and grouping (P<0.05); The expression of vimentin of both case and control groups satisfies sphericity test. The difference between the two groups is statistically significant (P<0.05), as well as that among different time points (P<0.001). There is interaction between time and grouping (P<0.05);3. The expression of Oct-4of both case and control groups does not satisfy sphericity test. After correction, the difference between the two groups is statistically significant (P<0.05), as well as that among different time points (P<0.001). There is interaction between time and grouping (P<0.05)Conclusion:1. TGF-β1can induce ACC-M cells to undergo morphological changes related to EMT;2. TGF-β1can up regulate the expression of mesenchymal cell related biomarkers N-cad, vimentin and down regulate epithelial cell related biomarkers E-cad to induce EMT in ACC-M cells; 3. TGF-β1can down regulate transcript factor Oct-4in ACC-M cells and may participate in the regulation of the proportion of different subpopulations of CSCs in SACC.
