Objective:(1)To analyze the expressions of CD44,CD133 in 41cases of tongue squamous carcinoma and 47cases of salivary adenoid cystic carcinoma. (2)To establish drug-resistant human Tca8113 cell line and Sacc cell line.and hypso-tumorigenetic human Tca cell line and Acc cell line.(3)To investigate the expressions of CD44,CD133 of the cell lines mentioned above and the characteristics of these cells.Method:(1)The expressions of CD44,CD133 were detected by immunohistochemistry staining in 41cases of Tca and 47cases of Acc.(2) Induce the DDP resistance of Tca8113 and Acc-M.DDP resistances of Tca8113 and Acc-M were induced by feeding these cells with DDP of single dose intermittently.Hypso-tumorigenesis cells were established by selecting cells of high multiplying ability through 3 times soft agar culture cloning.The index of drug-resistence was estimated by MTT.Use immuocytochemistry and FCM to investigate the differences of CD44,CD133 and apoptosis between induced cells and non-induced cells.(3)28 nude mice were divided into 6 groups randomly, and then the induced cells and non-induced cells were subcutaneous injected on the back of mice.Then the differences of the index mentioned above of transplanted tumors of each group were checked by using immunohistochemistry and TUNEL.Result:(1)There were CD44,CD133 positive cells in Tca and Acc tissues, but the expression had no significant differences between different histology grade of Tca and different histotype of Acc.(P>0.05).(2)Drug-resistant cells,Tca8113/DDP and Acc-M/DDP,were obtained, whose IC50 were 12.85μg and13.32μg,2.5 and 2.54 times of their parent cells respectively.Cells of higher multiplying ability,Acc8113/agar and Acc-M/agar, were obtained,whose percents of clone formation different significantly from their parent cells.(3)The expression of MRP had no significant difference between group Tca/agar and Tca8113 control group,the other differences beside the expression of MRP were significant(p<0.05).(4)The induced cells of Tca8113 and Acc-M grow faster and they can make large transplanted tumor than non-induced cells.Conclusion:(1)There are some CD44,CD133 positive cancer cells that have stem-like characteristics in Tca and Acc.But the expressions of CD44,CD133 are not concerned with histology grade and histotype.(2)The induced cells of Tca8113 and Acc-M have a stem cell-like ability,and have multiplication capability than non-induced cells.
