ObjectiveTo observe the effect of various concentration of testosterone on PTEN of human luteinized granulosa cells in vitro, so as to investigate the pathophysiological mechanism of testosterone on the apoptosis granulose cells(GCs) and provide new research ideas for polycystic ovary syndrome(PCOS).MethodsHuman luteinized granulosa cells from in vitro fertilization-embryo transfer (IVF-ET) are seperated from November2011to December2011in Xiangya hospital Reproductive Medicine Center of Central South University.They were differently divided into four groups after cultured48hours and then treated with testosterone at various concentration of, blank control group,10-7mol/L,10-6mol/L,10-5mol/L for24h without the serum. The expression of mRNA of PTEN of GCs were determined by RT-RCR. Apoptosis rate of GCs were detected by4,6-dianidino-z-phenylindole (DAPI) methods.Results1、 In the culture medium the monolayer cells appeared to be widespread and multiple interconnections between the cells after cultured48hours and were suitable for experiment in vitro2、 The expression levels of PTEN mRNAs significantly increased in the group treated with testosterone at10-5mol/L(P<0.05). Apoptosis rate was6.29±1.78%in the group of10-5mol/L and higher than the other groups(P<0.05).Conclusion 1、 PTEN expressed in human luteinized granulosa cells.Higher concentration of testosterone (10-5M) promote apoptosis of human luteinized granulosa cells.3、 Higher concentration testosterone (10-5M) can improve the expression of PTEN.The mechanism of apoptosis induced by testosterone in human luteinized granulosa cells might be related to apoptotic pathways mediateded by PTEN.
