ObjectiveObserving the dynamic changes of IGF-1and BGP in serum of IUGR rats, and the influence of puerarin intervention to the expression of IGF-1and BGP, discussing the possible mechanism of IUGR decreasing bone metabolism and the influence of puerarin intervention to it.Methods1. Establishment of pregnant rat model:Eighteen female and nine male healthy three-month SD rats were raised. Male and femal rats were put into a cage at the proportion of1:2, observed the femal rats by vaginal smear examination next day, see a large number of sperm for the first day of conception. Pregnant rats randomly divided into2groups:①Group of normal protein diet:Six pregnant rats in this group, were fed with normal protein diet during gestation period and lactation period, to establish the models of normal birth weight rats.②Group of low protein diet:Twelve pregnant rats in this group, were fed with low protein diet during gestation period and normal protein diet during lactation period to establish the models of IUGR rats.2. Groups of experiment and intervention measure to rat offspring:①Group of normal birth weight(Normal group):offsprings of mothers fed with normal protein diet;②Model group of IUGR(Model group): offsprings of mothers fed with low protein diet;③Puerarin intervation group of IUGR(Intervation group):offsprings of mothers fed with low protein diet,and intervened by puerarin(dose:50mg/kg.d) abdominal subcutaneous injection for14days from the8th day. Normal group and model group were intervened by sterile water to be compared. The three groups were breast feeding during lactation period and fed by normal protein diet after3weeks, with nature drink.3. Laboratory tests:body weight of pregnant rats and birth weight of neonatal rats were measured. Body weight of offsprings were measured regularly. Expressions of the serum of IGF-1and BGP were detected by ELISA at the age of1w,3w and8w respectively. 4. All data were statisticed by the SPSS17.0. Significant level was set at0.05.Conclusion:1、Comparison of weight:(1) Comparison of pregnant rats weight:Compared with group of low protein diet, the weight gain of pregnant rats in group of normal protein diet was statistically significant higher (P<0.05)(2) Comparison of birthweight:Birth weight of model group and intervation group were lower than normal group, with statistical significance (P<0.05)(3) Comparison of the change of offspring’s body weight:At the age of1w, offspring’s body weight of normal group was higher than model group and intervation group, with statistical significance (P<0.05), but with no difference between model group and intervation group(P>0.05). At the age of3w, the body weight of normal group was higher than model group and intervention group, with statistical significance(P<0.05),and model group was lower than intervation group(P<0.05). At the age of8w, there was no difference between normal group and intervation group(P>0.05), but all higher than model group(P<0.05).2、Comparison of the changes of serum IGF-1:At the age of1w, normal group was higher than model group and intervation group(P<0.05), but no statistical differences between model group and intervation group(P>0.05). At the age of3w, intervation group was higher than normal group and model group(P<0.05), and no statistical differences between normal group and model group(P>0.05). At the age of8w, there was no difference between the three groups (P>0.05).3、Comparison of the changes of serum BGP:At the age of1w, normal group was significantly higher than model group and intenvation group(P<0.05), no statistical differences between model group and intervation group(P>0.05). At the age of3w, normal group was lower than model group and intervation group(P<0.05), no statistical differences between model group and intervation group(P>0.05). At the age of8w, intenvation group was significantly higher than model group and normal group(P<0.05) but no statistical differences between normal group and model group(P>0.05).Conclusion1、Low protein diet in pregnancy can lead to poor weight gain in mid-late pregnancy and IUGR.2、At the age of1w, serum IGF-1and BGP of IUGR rats decreased. It is maybe one of the important mechanisms about the IUGR adulthood abnormal bone metabolism.3、Intervation of puerarin begin at the8th day of offspring rats can improve the level of serum IGF-1of IUGR rats, promote growth, and maybe improve the bone metabolism at8w.
