ObjectiveTo explore the effects of human beta-2defensin(hBD-2) through studying the expressions and locations of hBD-2in oral submucous fibrosis(OSF),oral lichen planus (OLP) and oral squamous cell carcinoma(OSCC).MethodsThe expression of hBD-2protein was examined by immuno-histochemistry method in30cases of OSF, including10cases of early, middle and late stage respectively,10normal oral mucous(NOM),15cases of oral squamous cell carcinoma (OSCC, grade1),15cases of oral lichen planus (OLP).The different positive cell rates of hBD-2in the five groups and immunohistochemical stains average gray values were compared by statistics analysis.Results1. There were positive expressions of hBD-2in NOM,OSF,OLP and OSCC. Brown staining was mainly located in epithelial cells, interstitial cells and membrane of the basal cells.2. The sort of positive cell rates and expression intensity of hBD-2among OSF,OLP, OSCC and normal oral mucous were OSCC>OSF(early stage)>OLP>NOM. The positive cell rate of OSCC was significantly higher than the others and there was statistically significant (P<0.05). There was no significant difference among OSF (early stage),OLP and OSCC(P>0.05),but the expressions of hBD-2in the middle stage of OSF was distinct higher3. The sort of the average gray values of hBD-2by large to small order was OSCC>OSF(middle stage)>OSF(late stage)>OLP>OSF(early stage)>NOM. There was significant difference between OSCC and NOM,OSF(early,middle and late stage),OLP (P<0.05). There was no significant difference between NOM and OSF (early stage),OLP (P>0.05)Conclusions1. There were differences in the expressions and locations of hBD-2among NOM,OSF,OLP and OSCC2. The differences in the expressions of hBD-2in three stages of OSF may imply there was relationship between hBD-2and the course of fibrosis.3. Further researches are needed whether hBD-2can be used as the reference index of diagnosis and prognosis of OSCC...
