BACKGROUND Pterygium is a common, benign, fibrovascul-ar,and infiltrative process of the corneal conjunctival junction of unk nown pathogenesis.Matrix metalloproteinases (MMPs) are a family of proteolytic enzymes active against all components of the extracellul-ar matrix,whose activity is specifically neutralized by tissue inhibitors of MMPs (TIMPs). Shanghai Synchrotron Radiation facility is advanced equipment in the world, three of the beam line could be used in the medical specimen.Research contents are microelement distribution a nd the three-dimensional structure and internal structure of pterygium. This study is designing experimental condition for pterygium and C onjunctival tissue in Synchrotron radiation echnology.Determine the microelement distribution the three dimensional structureand internal structure in pterygium tissue by using Synchrotron radiation.OBJECTIVE Design experimental condition for pterygium an dconjunctival tissue in Synchrotron radiation echnology to test pter-ygium relative trace elements content、distribution and characteristics of organizational structure.To investigate the role of MMP-2and M MP-9in thpathogenesis ofpterygia,and affording theory supporting for exploring new treatment method of pterygium.METHODS Design experimental condition for pterygium and conjunctival tissue in Synchrotron radiation echnology.Determin the distribution and its difference of Ca, Fe, Cu, Zn, Pb in pterygi-um tissue and control tissue by using Synchrotron radiation microbeam X-ray fluorescence analysis. Determine the three dimensional st-ruct-ureand internal structure of pterygium via X-ray imaging and Bio-m edical application and Small-angle X-ray scattering;The express-ions of MMP-2,MMP-9were examined in20primary pterygium,20conj-unctiva via immunohistochemistry in the fibroblasts.RESULTS1、When pterygium was in100g.L-1neutral formaldehyde fixed, making40μm frozen section,could be for good test on BL15U.when pterygium was in100g.L-1neutral formaldehyde fixed, through Str-ict dehydration,could be for good test on BL13W.2、The content of Ca, Cu, Pb in pterygium tissue is lower than in control tissue, The content of Zn, Fe in pterygium tissue is higher than in control tissue, the difference is significance (p<0.05).The microelement mostly is big shape and concentration collect in pter-ygium tissue, but in control tissue the distribution is dispersity.The three dimensional structure and internal structure of pterygium is h-oneycomb arrangement,not uniform.3、The positive rates of MMP-2and MMP-9in pterygium and control were75%,85%and40%,25%.There was statistical differ ence between pterygium andcontrol(p<0.05).The expressions of MMP-2and MMP-9were positively correlations.The coefficiency of MMP-2and MMP-9was0.585(p<0.007).CONCLUSIONS1、Control pterygium section thickness is key for BL15U test. Making40μm frozen section, playing on6μm mylar film is good for test on BL15U.Strict dehydration is necessary for test on BL13W.2、The relative microelement content、distribution and charact eristics of organizational structure of pterygium is researched via S-ynchrotron radiation,and have a good result.3、The positive rates of MMP-2and MMP-9in pterygium were higher than control, The expressions of MMP-2and MMP-9were positively correlations with synergistic effect.
