Objective:To investigate the effect of Icariin (ICA) and Simvastatin (SIM) with different dosage ratios on rabbits’radius fracture healing. And to provide the basis of animal experiment for treating fracture.Methods:The45health rabbits were made bilateral radial fracture as models and distributed to5groups at random, nine each. A:blank control group; B:BMP-2control group; C、D、E:experimenial groups. Group A and B were respectively given normal saline of20ml by gastric perfusion per1Kg. Group C,D, and E were respectively given ICA and SIM solution of15ml and5ml,10ml and10ml,5ml and15ml by gastric perfusion per1Kg. Group A,C,D, and E were injected normal saline of0.5ml in bone fracture location. Group B were injected BMP-2solution of0.5ml in bone fracture location. After2,4and8weeks,3rabbits in each group were randomly killed to evaluate whether there are significant differences among these groups according to general observation, X-ray examination, bone mineral density examination, histomorphometric examination and statistical analysis of data.Results:1. General observation and X-ray examination:The radius of all groups are healing as time passes. Group B and D is the earliest, followed by group C and E, group A is the latest in the growth and alteration of callus and recanalization of bone marrow cavity. There are no significant differences between group B and D, group C and E.2.bone mineral density examination:BMD of group A,B,C,D,E respectively is0.096±0.012g/cm2、0.150±0.008g/cm2、0.133±0.028g/cm2、0.152±0.013g/cm2,0.137±0.023g/cm2in the2nd week,0.108±0.015g/cm2、0.180±0.017g/cm2、0.153±0.038g/cm2、0.177±0.014g/cm2、0.150±0.055g/cm2in the4nd week,0.143±0.011g/cm2、0.095±0.025g/cm2,0.121±0.029g/cm2,0.100±0.008g/cm2、0.116±0.022g/cm2in the8nd week. There are significant differences among these groups in the2nd,4th,8th week (P<0.05).Significant differences can be found between each two groups except group B and D, group C and E through further compare (PBD、PCE>0.05, others P<0.05)3. biomechanical test:The maximum anti-contained of group A,B,C,D,E respectively is98.32±3.83N,139.10±23.59N,117.03±10.19N,141.68±8.44N、118.82±9.91N.There are significant differences among these groups in the8th week (P<0.05).Significant differences can be found between each two groups except group B and D, group C and E through further compare (PBD、PCE>0.05, others P<0.05)4. Histomorphometric examination:It’s indicated that group B and D is the earliest, followed by group C and E, group A is the latest in the growth of collagen fiber, chondrocyte and bone trabecula.There is no significant difference between group B and D,group C and E.Conclusions:ICA and SIM with different dosage ratios all can promote the radius fracture healing of rabbits when they are together used. When both of dosage is10mg/(kg· d), they can further promote healing.
