Objective:To evaluate the quality of Cyperi Rhizoma and establish new methods for its quality control, a series of studies including morphological indentification, microscopical identification of powder and TLC-identification were did. Besides, to determin the contents of a-cyperone in medicinal herb of Cyperi Rhizoma from different areas, the contents of total flavonoids, total sugar, polysaccharose were detected by UV.Methods:Ten batches of Cyperi Rhizoma from five different areas including Linyi, Taian, Weifang. Liangshan and Heze were collected. Morphological indentification and microscopical identification of power were employed to identify these Cyperi Rhizoma. TLC identification of a-cyperone and digitoflavone were used. The contents of water, ash, extract, volatile oil in Cyperi Rhizoma were determined. The contents of a-cyperone, toal sugar, polysaccharose, general flavone were analyzed by HPLC and UV.Results:1. No obvious differences were observed in appearance character and powder microstructure of Cyperi Rhizoma from different regions.2. The specificites of TLC identification methods had been definited. The result of TLC identification of a-cyperone and Luteolin in Cyperi Rhizoma from different areas showed that the same tinctorial blotches in the right position were revealed.3. The contents of water in Cyperi Rhizoma from different regions were6.195%~8.370%.The contents of total ash were1.74%~3.12%and sour insoluble ash were0.29~0.78%.4. The contents of Extract thin alcohol in Cyperi Rhizoma were changed from16.02%to24.02%.5. The contents of volatile oil in Cyperi Rhizoma from five areas were all above1%and the highest content is Weifang.6. HPLC was used to determine the contents of α-cyperone and the result showed that the contents of a-cyperone were0.0677%~0.1244%. This method is scientific and easy to operation.7. By a series of experiments, we established new methods by UV to detect the contents of total sugar, polysaccharose and general flavone. The content of general flavone was hightest in Linyi and the others were changed from1.12%to1.82%. For sugar, the content is obvious different from area to area, the content of total sugar and polysaccharose in Weifang is hightest. The others were17.53%~38.66% and1.88%~20.26%.Conclusion:According to the study of Cyperi Rhizoma from different areas, we have established the morphological and powder microscopical characters. Besides, new TLC identification method of morphological was established. We also built new method to determine the contents of total sugar, polysaccharose and general flavone. All these methods provide the scientific basis for quality standards of Cyperi Rhizoma.
