Part1Experimental studyObjective: Study the influence of cantharidin of Chinese medicine extract to tumorproliferation on HEL cells, and detector the expression of JAK2gene, in order to know theinfluence of cantharidin to clarify anti-tumor effect, and expounds the relevance betweenits mechanism and the expression of JAK2gene.Methods: By MTT method to affirm theinhibition of proliferation of cantharidin to the HEL cell.The RT-PCR half quantitativemethod to detect different concentrations of cantharidin HEL cell lines JAK2geneexpression of the influence of the situation.Result: Cantharidin inhibits HEL cellproliferation, significant time and dose effect; JAK2gene expression of the quantity is allhas significant difference between blank group and dose of cantharidin group, blank groupand between high doses of cantharidin between high doses of cantharidin and betweengroups（P<0.01）;Both dose of cantharidin group and high doses of cantharidin group canreduce the relative expression of JAK2gene(P <0.01); The relative expression of JAK2gene of high dose group is less than the dose group (P <0.01).Conclution: The thing thatcantharidin can inhibit proliferation of HEL cell lines relevanted to it can reduce the JAK2gene expression.Part2Clinical researchObjective: Research compound BanMao capsule with western medicine on theclinical Effectiveness in patients with PV.Methods:40patients with PV were sorted to two groups of experimental group and control group in1:1proportion and random.Controlgroup is given routine western medicine,experiment group is given routine westernmedicine combined with compound BanMao capsule,Observe the clinical effectivenessafter3months.Results: The difference of Age (P=0.160), gender (P=0.749) and duration(P=0.778) between experimental group and control group there were no significantdifferences in comparable have no statistical sense; Two groups of hemoglobin and threedepartment are both no significant differences (P>0.05),and are comparable..Aftertreatment, the difference about the total curative effect comparison, the total effective rateof the experiment group were90%and control group is84%, the difference was notstatistically significant (P=0.179). Syndrome of two groups is (P=0.037), total scoresyndrome (P=0.009), all has a statistically significant difference. Hemoglobin content ofTwo groups is (P=0.037), platelet numerical comparison (P=0.045), white numericalcomparison (P=0.001), numerical comparison red blood cells (P=0.006), all have astatistically significant difference.Conclution: Compound BanMao capsule combined withwestern medicine can improve the curative effect in PV patients.
