Background&Objective:The gastric cancer is the most common tumor in our country, the chemotherapy is still the major means of comprehensive treatment gastric cancer. But in recent years the clinical treatment of traditional antitumor drugs not sensitive patients more and more, the serious influence the patient’s survival, the tumor cells to chemotherapy appear very pressing against. A lot of research shows GRP78, MCL-1in a wide variety of tumor tissue has high expression state, and it is thought it and these tumors, development and resistance to chemotherapy treatment and so on. This research mainly to clinical entity specimens for example, combined with the culturing in gastric adenocarcinoma cell lines, this paper discusses GRP78, MCL-1protein expression and cancer of the gastric and the clinical and pathological features of correlation in the gastric cancer occurrence, development of the role of MCL-1protein in the regulation of endoplasmic reticulum stress induced cancer cells the role of the gastric adenocarcinoma in the apoptosis, so as to provide clinical chemical medication gastric cancer effective targets..Methods:Tissue microarrays containing gastric cancer tissue and gastric normal tissue were prepared and the expression of GRP78and MCL-1protein were detected by immunohistochemistry. Western blot was used to test the expression of GRP78and MCL-1protein in BGC823, SGC7901gastric cancer cell lines induced by3μmol/L tunicamycin in different periods.Results:Immunohistochemistry results show that:the gastric cancer tissues and gastric normal tissue are GRP78and MCL-1protein expression, located in the cytoplasm, shading pale yellow to dark brown, but MCL-1protein expression is relatively weak. Statistics show GRP78in gastric cancer tissue expression in the index of5.80+/-3.45, the gastric normal tissue for3.20+/-1.70, both have obvious difference (t=3.259, p=0.002). MCL-1protein in the gastric cancer tissue, gastric normal tissue positive expression rate were86.49%(124/148),60.00%(12/20), and also a statistically significant difference (the chi-square=6.464, P=0.011). MCL-1protein expression and is associated with lymph node metastasis related, and GRP78protein seemingly; patients and the age, sex, tumor size, infiltration depth, etc are no correlation, but and gastric cancer cell differentiation degree are negatively. Western blot display gastric adenocarcinoma cell line SGC-7901hare tunicamycin induction endoplasmic reticulum the condition of stress MCL-1the protein expression with time points in extended raised the trend, and BGC-823-1protein MCL cell lines are in different time points high expression state, no significant increase trend.Conclusion:GRP78and MCL-1was highly expressed in gastric adenocarcinoma, and correlated with gastric carcinogenesis, the degree of differentiation of gastric carcinoma cell, MCL-1involved lymph node metastasis,, but both GRP78and MCL-1not associated with the patient’s age, sex, tumor size and tumor infiltration depth. MCL-1protein and gastric adenocarcinoma cells to the endoplasmic reticulum stress tolerance of apoptosis related, tip GRP78, MCL-1genes or protein can be used as the management of gastric cancer effective targets.
