ObjectiveLaryngeal squamous cell carcinoma occurs laryngeal epithelial malignant tumor,is one of the most common malignant tumors in otolaryngology head and necksurgery. Laryngeal cancer suppressor gene RASSF1A expression and laryngealsquamous cell carcinoma in recent years domestic rare. RASSF1A gene interventsthe cell cycle, inhibits cell growth, promote cell apoptosis and suppressestumorigenesis. The gene promoter inactivation occurred after hypermethylation,leading to disorder of cell cycle and cancerous. The study observed RASSF1Aexpression in laryngeal squamous cell carcinoma tissue, discussed on the relationshipbetween RASSF1A expression and occurrence and progression of laryngealsquamous cell carcinoma, in order to make it become a new diagnostic andprognostic evaluation index, improve the early diagnosis rate and cure rate of cancer.Laryngeal keratosis, premalignant lesions of laryngeal cancer, is a mucosaepithelial growth, maturation abnormalities as well as mucosal hyperkeratosis aproliferative diseases. The hypermethylation and expression of RASSF1A studiesrarely in laryngeal premalignant lesions. The study observed RASSF1A expression inlaryngeal keratosis and gave a preliminary study on correlation between them.MethodsWe collected the specimens of polyp of vocal cord, keratosis of larynx andlaryngeal squamous cell carcinoma in Otolaryngology-Head and neck surgery of Anhui Provincial Hospital from2009-2011and took normal vocal fold mucosa ofCadaveric head.Immunohistochemical methods were used to detected the expressionof RASSF1A protein in these tissue. Diffuse distribution of brown granules in the cellcytoplasm was positive result and the nucleus was not coloring. Calculated thepercentage of positive cells of every section,then took the average value±standarddeviation(x±s).Using SPSS17.0statistical analysis software to analyze the data, P<0.05for the difference was statistically significant.ResultsRASSF1A in laryngeal squamous cell carcinoma and laryngeal keratosis tissuesbelow the vocal cord polyps and normal laryngeal mucosa; Laryngeal squamous cellcarcinoma and polyp of vocal cord, laryngeal squamous cell carcinoma and normallaryngeal mucosa and squamous cell carcinoma of the larynx and laryngeal keratosiswere compared, the difference of RASSF1A expression were statistically significant(P <0.05).Keratosis of larynx and vocal cord polyp, keratosis of larynx and laryngealmucosa of normal and all types of laryngeal keratosis were compared, the differenceof RASSF1A expression was not significant (P>0.05).RASSF1A expression in the clinical stage from I to IV of laryngeal squamouscell carcinoma gradually declined. During the clinical stage, difference has statisticssignificance (P <0.05). In high differentiated squamous cell carcinoma, moderatelydifferentiated squamous and poorly differentiated squamous, the difference ofRASSF1A expression was not significant,nor the differences between age groups(P>0.05).Conclusion1. In laryngeal squamous cell carcinoma tissue,RASSFlA expression wassignificantly lower than that in normal tissue and vocal cord polyp. This indicatedthe relationship between the reduced expression of RASSFlA and laryngeal squamous cell cancer.2. The expression of RASSFlA had no relationship with age and tissuedifferentiation, but had relationship with the clinical stage. RASSFlA proteinexpressed different degrees in different clinical stage of laryngeal carcinoma.RASSFlA protein expressed higher in the early stage than in the later clinicalstage. This suggests that RASSFlA expression may be associated with tumorinvasion and metastasis.3. The RASSF1A gene may be involved in laryngeal squamous cell carcinoma inearly stage. Therefore we can use this feature as a early diagnosis indexes oflaryngeal squamous cell carcinoma, and for the identification of premalignantlesions4. The detected of RASSFlA protein expression may contribute to the screening ofRASSFlA gene abnormalities in the crowd, and improve the early diagnosis levelof laryngeal squamous cell cancer.
