Objective:To investigate the effect of ketamine on the capabilities of memoryand learning and AchE,ChAT of hippocampus in neonatal rats.Methods:80aged7days SD rats of both sexes weighing12~18g wererandomly divided into group C、 K1、 K2、 K3,20per group. Group C wasintraperitoneally injected with2ml physiological saline, group K1was intraperitoneallyinjected with80mg/kg ketamine once, group K2was administrated with80mg/kgketamine once a day for2days, group K3was administrated with80mg/kg ketamineonce a day for3days.10rats were killed each group the next day after the last injectionand isolated the hippocampus for determination of ChAT and AchE. After testing thebehavior in21th by Morris water maze(MWM), the rest of rats were killed and isolatedthe hippocampus for determination of ChAT and AchE.SPSS13.0was applied toanalyze the numbers of ChAT, AchE in hippocampus in each group and the changes oflearning and memory in MWM.Results:MWM: There was no significant difference between group C and K1and groupK2and K3(P>0.05), the latency period were longer and the frequency tocross the plat were less in group K2,K3than group C,K1(P<0.05). The results ofenzymes: the8th, there were significant difference (P<0.05); The27th, there was no significant difference between group C and K1and group K2and K3(P>0.05). Therewas significant difference between group K1,C and group K2,K3(P<0.05).Conclusion:Administration of80mg/kg ketamine can change the ChAT, AchE inhippocampus of aged7days SD rats. Administration of80mg/kg ketamine once maychange the ChAT, AchE in hippocampus in24hours. Repeated injection can change theChAT, AchE in24hours and in the21day, and administration of ketamine differenttimes arise different results. Injection once can not impair learning and memory abilityin the21day, and repeated injection can impair learning and memory ability in the21day, but compared with administration ketamine2times, administration3times can notresult in significant change in the ChAT, AchE and learning and memory ability. Thecholinergic nervous system of hippocampus is related to the spatial learning andmemory behavior of neonatal rats, the change of learning and memory ability impairedby ketamine may be related to the ChAT, AchE in hippocampus.
