purpose To detect the expression of TRAIL protein and its receptor of DR5, DcRl in patients with lesions of CA, and know about the possible effects and meanings of TRAIL-mediated cell apoptosis in the onset and recurrence of CA.Materials and Methods Using the Immunohistochemical peroxidase labeled streptavidin avidin method (S-P) to detect30cases of primary CA patients’skin lesions,30cases of recurrent C A patients’skin lesions expression of TRAIL protein and its receptor DR5, DcRl in skin.and30cases of normal skin as control group. The results were determined by semi-quantities test,and the statistical analysis was managed by SPSS17.0for windows software.Results1、TRAIL protein’s immunoreactive product is a brown granular calm, which located in the cytoplasm and membrane. The cells were mainly distributed in the prickle cell layer. The positive expression rate of TRAIL in control group, onset group and the recurrence group were91.67%,83.33%and80.00%, which showed a decreasing trend. The expression intensity between control group and CA group differs. The control group group’s is higher than the onset and recurrent groups (p<0.05), while between the onset and recurrent groups was not statistically significant (p>0.05).2、DR5protein’s positive staining is in the cytoplasm. more than pale yellow or brown, patchy or diffuse expression. The positive cells were mainly distributed in the basal cell layer and the prickle cell layer.DR5protein’s positive expression rate in the control group, onset group, recurrence group of CA is53.33%,83.33%and93.33%, which showed a increasing trend. The expression intensity differences in the control group is lower than CA groups,which was statistically significant (P<0.05); while between the onset and recurrent groups was not statistically significant (p>0.05).3、DcRl protein’s positive staining is in the cytoplasm. more than pale yellow or brown, patchy or diffuse expression. The positive cells were mainly distributed in the prickle cell layer. DcR1protein positive expression rate of the control group, onset group and the recurrence group were100%,57.50%,40.00%. Showed a decreasing trend. The expression intensity difference between any two groups were statistically significant (P<0.05).Conclusion1、The abnormal low expression of TRAIL in CA skin lesions may be related to the development of CA.2、TRAIL expression in the onset and recurrent groups showed no difference, suggesting that TRAIL may not be the main involved in the recurrence of CA.3、Upregulation of DR5in CA, may provide the basis for clinical treatment of exogenous TRAIL.4、Downregulation of DcRl in the recurrence group, lower than the onset group, suggesting that DcR1may be one of the reasons of the HPV-infected cells to evade immune surveillance, resulting in recurrence of CA.5、Downregulation of DcRl, combined with the upregulation of DR5in CA, may explain the mechanism of exogenous TRAIL-targeted destruction of infected cells.
