Objective To study the expression of hypoxia inducible factor-1α and H.pyloriinfection in gastric cancer. And explore the relationship among expression of HIF-1αand H.pylori infection during the proceed and development of gastric cancer.Methods60patients of gastric cancer and36patients of chronic gastritis were usedfor ELISA to determine the IgG antibody of H.pylori. The expression of HIF-1α weredetected by immunohistochemistry in60patients of gastric cancer and20patients ofchronic gastritis. And the correlation among HIF-1α expression, H pylori infectionand the progress and development of gastric cancer were analysis by SPSS17.0software. The expression of HIF-1αwere detected by western blot after MGC-803cellcoculturing with H.pylori at different time0h,6h,12h,24h,48h.Result1) The positive ratio of H.pylori infection was86.1%(31/36) in chronicgastritis was higher than66.7%(40/60) in gastric cancer. And there were significantdifference between them (p<0.05);The positive ratio of H.pylori infection was72.7%(8/11)in early gastric cancer which was lower than that in advanced gastric cancerwith a ratio of65.3%(32/49). And there were significant difference between them (p﹥0.05).2) The positive ratio of HIF-1α were40.0%(8/20),80.0%(48/60)in chronicgastritis, and gastric cancer;54.5%(6/11),85.7%(42/49)in early gastric cancerand advanced gastric cancer, respectively. And there were significant differencebetween them, respectively (p<0.05).3) The positive ratio of HIF-1α was80.0%(48/60)in gastric cancer. And44.4%(4/9),82.1%(23/28),91.3%(21/23)in high-differentiated, middle-differentiatedand low-differentiated gastric cancer, respectively. And there were significantdifference among them (p<0.05). 4) The exression of HIF-1αwere upregulated after induced by H.pyloricompared to uninfected group in MGC-803(P <0.05). The expression of HIF-1αweresignicant upregulated after12h H.pylori infected compared to0h,6h. And there weresignificant difference among them (p<0.05).Conclusions1) The expression of HIF-1αwere higher in gastric cancer than chronic gastritis,in advanced gastric cancer higher than early gastric cancer, in low differentiated andundifferentiated higher than middle-high differentiated gastric cancer. There werecorrelation among expression of HIF-1α and the progress and development of gastriccancer.2) There were correlation between H.pylori infection and the expression ofHIF-1α. And the hypoxia environment induced by H.pylori infection leaded toupregulaiton of HIF-1αin gastric cancer may be one of mechanism involving in theprogress and development of gastric cancer.
