OBJECTIVE To optimize the extracting process of alkaloid from Nelumbonucifera leaves.To establish NAFLM in mice by high fat feeding and carbontetrachloride. To investigate whether alkaloid in lotus leaf had therapeutic effect onnonalcoholic fatty liver and its possible mechanism in mice.METHODS (1)The spectrophotometry was used to determine the alkaloids.Theextracting process of alkaloid was optimized by L16(44) to observe the effect ofextraction time,extraction temperature, solid to liquid ratio, and ethanolconcentration, respectively.(2)Kunming mice were randomly divided into threegroups: control, high fat, and high fat plus carbon tetrachloride. Liver morphologicaland pathological changes were observed at2,4,6and8weeks. Serum levels of totalcholesterol (TC), triglyceride (TG), alanine transaminase (ALT) and aspartatetransaminase (AST) and hepatic TC, TG changes were determined.(3)48mice weredivided into four groups, normal group,model group, essentiale group, alkaloid inlotus leaf group(100mg/kg). The normal group fed with basic animal feeds.Nonalcoholic fatty liver model mice were successfully established by high fat diet for60days. The experimental mice were treated with alkaloid in lotus leaf for42daysfrom the14th days. The therapeutic effects of alkaloid in lotus leaf on nonalcoholicfatty liver and histopathological changes of mice liver were observed.RESULTS (1)The optimum extraction parameters were extraction time2hour,ext raction temperation90℃, solid to liquid ratio1:15,(2) In high fat plus carbontetrachloride group, inflammatory cell infiltration appeared in the mice liver after2weeks. There were a small amount of lipid droplets at4weeks, and steatosis emergedafter6weeks. Hepatic fibrosis began to appear at8weeks. The serum levels of totalTC, TG, ALT and AST, and hepatic TC, TG were significantly advanced in high fat plus carbon tetrachloride group compared with control group at8weeks (P <0.05orP <0.01).(3)After treatment with alkaloid in lotus leaf, the mice serum levels oftotal cholesterol(TC), triglyceride(TG), alanine transaminase(ALT) and aspartatetransaminase(AST) were significantly decreased as compared with the nonalcoholicfatty liver model group(P<0.05or P<0.01).CONCLUSION:(1)Lotus leaf extract the best condition of alkaloids,temperature of90℃, pH value of2.5, extraction2h, solid-liquid ratio is so. Get oflotus leaf alkaloids, the purity of73.4%.(2)Lotus leaf alkaloids can lower blood andliver tissue of TC, TG, ALT, AST content, effective slow nonalcoholic fatty liverdisease the occurrence and development of the disease.(3)Lotus leaf alkaloids lowerblood and liver tissue of TC, TG is better than good role to the essentiale...
