Objective:To study the distributions of N-Methyl-D-aspartate receptor subunit1(NMDAR1) inthe adult brain and compare the distributions in different regions of the adult brain.With these data, the role of NMDAR1and the mechanisms of alcohol addiction anddrug addiction will be studied better in future. Furthermore, these data are also helpfulto the location in therapies for drug addiction by surgery and Deep Brain Stimulation.Methods:Five adult brains were collected and made to tissue chips. Detect the distributions ofNMDAR1in different regions of the adult brain by immunohistochemistry, comparethe NMDAR1expression levels in different brain regions between male and femle.Results:Immunohistochemistry results showed NMDAR1distributes in all parts of the adultbrain. NMDAR1positive cells were identified by brown staining cell membrance andlight brown staining cytoplasm, and sometimes nucleoli can be seen too; theexpression levels of NMDAR1are various in different brain regions(P<0.05). Theexpression levels are showed from higher to lower: Hippocampus, Cerebral cortex,Corpus striatum, Thalamus, Brainstem, Cerebellum; the expression levevl ofNMDAR1in dentate gyrus is higher than that in Hippocampus CA1,CA2,CA3regions (P<0.05). However, There is no significant difference between the expressionlevels of NMDAR1in Hippocampus CA1, CA2, CA3regions (P>0.05).Conclusions:1. NMDAR1distributes widely in the adult brain,the different brain regions whichhave higher express level of NMDAR1to lower are as follows: Hippocampus,Cerebral cortex, Corpus striatum, Thalamus, Brainstem, Cerebellum; 2. The expression level of NMDAR1in dentate gyrus is the highest in Hippocampus,no significant difference has been detected in CA1,CA2,CA3regions.
