Objective:To explore the effects of the activity astrocyte EGFR-JAK1/STAT3SignalingPathway in Rats with Intracerebral Hemorrhage.Methods:The male SD rats were randomized into3groups:GSTgroup(20)、ICH group(20)、DSMOgroup(20)、. The three groups used to make cerebral hemorrhagemodels by injecting VII collagenase into the caudate nucleus, The rats in GSTgroups were injected with three different drugs50mg/kg/d once a day respectivelyuntil they were put to death. The rats in DSMO groups were injected with DMSOsolution once a day; The rats in ICH groups were injected with saline once a dayrespectively until they were put to death. In1d,7d,14d,28d after the operation, allthe rats’ neurological function were graded. Five rats were randomly selected tosacrifice in each time point in each group,Meanwhile, immunohistochemistry wasused to detect the expressions of GFAP and EGFR on astrocytes in the brain tissue,and westernblot was used to detect the expressions of p-JAK1and p-STAT3.Results:Neurological function scores showed no significant differences in motor functionrecovery between Group DMSO and Group ICH.while in Group GST motor functionrecovery is better than the former two groups (P<0.05), Results ofimmunohistochemical indicated:.The expression of GFAP and EGFR were increasedafter ICH, Compared with Group DMSO and Group ICH, in Group GST, there was astatistical decline.(P<0.05). Results of Western blot indicated:in1th.14th.28thafterICH,the three group p-JAK1/p-STAT3-protein expression was no significant change,but in7th.after ICH, p-JAK1and p-STAT3protein expressions were increased,in group GST p-JAK1/p-STAT3protein expressions were less than Group DMSO andGroup ICH, which had significant differences(P<0.05).The expression of GFAP、EGFR,p-JAK1/p-STAT3protein lerels were not affected by DMSO solution used forGST.Conclusion:1. The expression EGFR were increased after astrocytes were activited; In Ratswith Intracerebral Hemorrhage. The expression p-JAK1/p-STAT3. were increasedtoo.2. Inhibit the expression EGFR that can level down The expressionp-JAK1/p-STAT3.3.EGFR-JAK1/STAT3signaling pathway may play an important role in theactivation of astrocytes in rats with intracerebral hemorrhage.
