Objective:Parkinson’s disease (Parkinson’s disease, PD) is a neurodegenerative diseases, in recent years, a large number of studies show that oxidatie stress for neurodegenerative diseases events play a very important role. Therefore, exploring the from natural products or find the antioxidant compounds and clarify mechanism, to prevent such the happening of the disease has the important practical significance. Astragaloside Ⅳ with antioxidant, scavenging free radicals, anti-inflammatory and so on many kinds of biological activities, but is it effective Astragaloside Ⅳ inhibition caused by oxidative stress damage neurons and has not yet been reported. Through the establishment of PC12cells and SH-SY5Y cells in vitro model oxidative stress, this paper discusses a PC12cells of Astragaloside Ⅳ and SH-SY5Y cells oxidative stress the protective effects of damage and mechanism, and effective prevention and control for neurodegenerative diseases provide beneficial experiment data.To investigate the effect of astragaioside Ⅳ on PC12cells injury induced by H2O2and its underlying mechanism. Method:The viability of PC12cell and SH-SY5Y cell induced by H2O2was detected by MTT assay. The status of nucleic acid in cell was observed by Hoechst33258staining. Apoptosis of PCI2cells,Reactive oxygen species(ROS) and cell cycle was measured by flow cytometry.The expression of Cyclin D1,Cyclin A,bax/bcl-2,Phospho-p38and T-p38,Phospho-JNK,Phospho-ERK and T-ERK were examined by Western blot.Results:The model on oxidative stress of PC12cell induced by H2O2was constructed successfully. The cell viability of PC12injured by H2O2was rescued by astragaioside Ⅳ, which could inhibit apoptosis of PCI2cells and reduce ROS and increase percentage of G1-phase in cell cycle. Mechanism research shows that:Astragaioside Ⅳ could rescue the descending expression of Cyclin D1and the increasing expression of Phospho-p38induced by H2O2.Astragaioside Ⅳ could rescue the expression of bax/bcl-2and a-syn,and the increasing expression of Phospho-p38induced by H2O2.Conclusion:Astragaioside Ⅳ protects the PC12cell from oxidative stress induced by H2O2via stimulating the expression of Cyclin D1and inhibiting active Phospho-p38. The present data suggested that Cyclin D1and Phospho-p38might be a target of clinical therapy via antioxidation.Astragaioside Ⅳ protects the SH-SY5Y cell from oxidative stress induced by H2O2via recovering the expression of bax/bcl-2and a-syn and inhibiting active Phospho-p38.It is a insight into prevention of neurodegenerative disease.
