Objective:To study the correlation between plasma brain-derived neurotrophic factor (BDNF) level and BDNF gene polymorphism (G196A) and the age of onset, sex, family history, suicidal behavior and antidepressants response of depressed patients; Explore the correlation between BDNF gene polymorphism (G196A) and depression gender, age of onset, family history, suicidal behavior and antidepressants response to treatment; Explore the correlation between plasma BDNF level and G196A gene polymorphism.Methods:A total of125cases of patients with depression collection, the Hamilton depression rating scale (HAMD24), and Hamilton anxiety scale (HAMA) and beck depression from review (BID) rating. Acquisition of patients and healthy controls were blood samples, with PCR amplification genomic DNA. The PCR product is used for enzyme cut and electrophoresis. Use the human brain source neurotrophic factor enzyme league immune detection to detect plasma BDNF level respectively.4,8,12weeks after treatment, each scale is measured again and blood plasma BDNF level is tested again.Results:(1) before treatment, the plasma BDNF expression level of patients is lower than the control group (case group946.672±297.075pg/ml, and the control group was1187.360±236.026pg/ml, P=0.019). the plasma BDNF expression level of patients with a history of attempted suicide is lower than none suicide group (778.882±230.709pg/ml;999.900±297.584pg/ml, P=0.024). After treatment, BDNF expression level has risen.4,8,12weeks after treatment, observe dynamically lower, and found those one is higher than baseline level. Compared with controls,4,8weeks after treatment, the tendencies of plasma BDNF level are increasing, but they are lower than normal control group, however,12weeks after treatment, the plasma BDNF level is higher than normal control group.(2) There is no clear correlation between plasma BDNF level, HAMD scale score and depression duration (r=0.099, P=0.273; r=0.003, P=0.975; r=0.126, P=0.88). There is no clear correlation between plasma BDNF level, gender, family history, age of onset (r=0.071, P=0.432; r=0.103, P=0.254; r=0.051, P=0.570).But the suicidal idea and suicidal behavior and a significant relationship with plasma BDNF level (r=0.250, P=0.005; r=0.298, P=0.000).(3) Genetic polymorphism alleles, genotype distribution frequency comparison between depression group and control group G196A:three genotype and A, G two allele frequency distribution has no significant difference; Gender, age, family history, suicidal behavior has no significant impact on genotype distribution.(4) BDNF gene polymorphism (G196A) of the same kind of genetic type and the expression level of plasma BDNF has significant difference between case group and control group,but the former is lower than the latter.(5) BDNF gene polymorphism (G196A) the genotype and the plasma BDNF level has no significant correlation.Conclusion:1. The group of patients with a history of attempted suicide has lower plasma BDNF levels than none suicide group and control; There is significant relationship between Plasma BDNF level and the suicide suicidal behavior. As time went on, the plasma BDNF level after antidepressant medication treatment is increasing. Considering it may be a biologic sign of depression. The plasma BDNF level may be a biologic sign of depression. Low levels of plasma BDNF could be a danger signal of suicidal ideation and attempted suicide of depressed patients.2. BDNF gene polymorphism G196A is related to the reaction of antidepressants, but it has no significant effect on A and G alleles.
