The Effect Of Lumican On The Proliferation And Invasion Of Human Lung Adenocarcinoma Cell Line A549in Vitro

Obsjective: To evaluate the effect of overexpression of Lumican on theproliferation and invasion of lung Adenocarcinoma cell line A549. Method:The stable cell lines were obtained by puromycin screening to select theA549cells that was infected by recombinant lentiviral vector carryinghuman Lumican gene. The mRNA expression of Lumican gene in the A549cell group，the empty vector group and the Lumican infection group weredetected with Real-Time PCR respectively． The protein expression ofLumican gene in the the three groups were detected with Western blottingmethod．The MTT assay was used to investigate Lumican gene，s effect onproliferation of the A549cells，meanwhile the transwell method was used todetect it，s effect on invasion of the A549cells in vitro.Results:(1)Theexpression level of Lumican mRNA in the each group was significantlydifferent（χ2=21.60， P <0.01）.The expression level of mRNA inLumican infection group was significantly higher than that in the other twogroups.The expression level of Lumican protein in Lumican infection group was higher than that in the other two groups（F=102.86，P <0.0001）.(2)There were no significant difference in OD values detected by MTT assayamong the three groups at each time point after induction（P>0.05）.(3)The number of transfected cells that passed the well in the each group wassignificantly different（χ2=23.49， P <0.01）. The number of transfectedcells that passed the well in Lumican infection group was significantlyhigher than that in the other two groups（F=73.92，P <0.001）. Conclusion:The A549cell lines that stabely expressing Lumican is successfully selectedin the study.Lumican gene is not involved in proliferation of A549cells， meanwhile it promotes the ability of invasion of the A549cells invitro.
