Experimental Study Of Effect Of Fibrin Glue Associated With Antisense To PCNA On Preventing Restenosis Of Vein Graft

Objective:To approach the effect and the mechanism of fibrin glue associated with antisense to PCNA on preventing restenosis of Vein Graft.Methods:Right jugular vein to common carotid artery interposition grafting was Performed in30male New Zealand white rabbits(2.5-3.0kg) and the animals were divided into three groups:non—support group(NS group;n=10); fibrin glue group (FS group;n=10) and fibrin glue+antisense group (AS/FS group;n=10). Commercially available fibrin glue and fibrin glue mixing with adenovirus expressing the antisense oligocleotides to PCNA was applied separately around vein grafts.At28th days after implantation, the grafts were harvested. Pathological analyses of the vein graft intimal and media thickness and area.RT—PCR analyses of PCNA gene expression.Results:28days after the operation, the thickness of intimal,media and the areas of intimal, media in AS/FS group (0.550±0.050) mm2.(0.437±0.054) mm2、(36.68±4.84) μm、(26.50±4.15)μm had significantly decrease compared with NS group (1.025±0.103) mm2、(0.944±0.084) mm2、(76.87±4.32)μm、(55.98±4.33) μm and FS group (0.743±0.052) mm2、(0.643±0.056) mm2(50.28±4.69) μm、(36.46±4.36)μm (P<0.05);RT—PCR analyses showed that the mRNA of PCNA was decreased in AS/FS group compared with NS group and FS group(P<0.05).Conclusions:The expression of PCNA in vein grafts can be inhibited by adventitial Delivery of antisense to PCNA. The fibrin glue associated with antisense has synergistic effect on reducing neointimal and medial thiekness of vein grafts. The fibrin glue associated with antisense to PCNA can preventrestenosis of vein graft.
