The development of modern society for prople’s life provides a rich material guarantee, at the same time human reproductive health by environmental change and the quickening pace of life is deteriorating.Less refined weak sperm and no refined disease is the main cause of male infertility in the obviously rising trend. We can ues stem cells to get the root of male infertility treatment.Embryonic stem cells (ESCs) can differentiate to the germ cells on the specific condition influence, but because of its source,tumor transplant in the body, immune rejection and social ethics limits its application in clinical, BMSCs (bone marrow mesenchymal stem cells) are un-hemopoietic tissue mesenchymal stem cells in bone mesenchymal, they can differentiate to adipose cells, bone cells, cartilage cells, nerve cells, muscle cells, kidney cells and other somatic cell,but if BMSCs can differentiate to the germ cells have rare study reports at present. So, this experimental groups were abdominally injected with different concentrations of busulfan (15、20、30、40、60mg/kg), and the sperm by random sampling every10d was observed until infertility status appeared such as low number and weak characteristics. The appropriate injection volume of busulfan was selected through testing the effects of busulfan on sperm concentration, vitality, malformation rate and the internal structure of seminiferous tubules.At the same time,in vitro culure of bone marrow stromal stem cells,immunohistochemical detection of BMSCs antigen CD44,CD106and generative cells surface antigen c-kit gene expression,transplantate BMSCs to the no-refined disease rat, detection testicular tissue cell structure, immunohistochemical testes the CD44,CD106,c-kit gene expression in the testicular tissue cells after30days.As the result, the rats within10d all death after injection busulfan once,dose20mg/kg(weight)for10d intraperitoneal injection busulfan are all survived. The internal structure of seminiferous tubules after injection of busulfan20mg/kg30days for the single seminiferous cell wall, and the control group testicular have a large number of cavity reproductive cells.From the rat femoral cavity separated the cultivation of bone marrow stromal stem cells, immunohistochemical results are shown that CD44(+),CD106(+) and c-kit(-).Injected the rBMSCs in the rat testes, there are lot of new cells in the testes after injected30d,and immunohistochemical shown c-kit(+).Results shown that:BMSCs can differentiate germ cells in the testes, and repair the damaged infertility in testis convoluted tubule of rats,For human bone marrow stromal autologous stem cells transplantation in the treatment of less refined disease application to provide theoretical and experimental basis.
