| Background and aimsIn recent years, with the development of social industrialization, the incidences ofallergic diseases are on the rise around the world, among which allergic skin diseasesare ranked on the top. Thus they have attracted more and more researchers to explorehow to apply effective therapies to prevent and cure them, which becomes one of thefocuses in the medical field.Avoiding contact with allergens, medication (antihistamine and corticosteroid),and specific immunotherapy are currently the main means of treatment for allergic skindiseases. But whatever methods we use, we cannot change the fact that the overall curerate of allergic skin diseases is relatively low, and the patients’ life quality is stillunsatisfactory. As a result, chemoprophylaxis against allergic diseases is becoming afocus of research in recent years. Chemoprevention is a method applying naturalproducts or chemical substances to reverse, inhibit, and prevent diseases. Now themost promising method of antianaphylaxis exists in the natural agents of food.Compared to artificial chemical substances, natural ones are safer, more economic, andmore acceptable.Epidemiological evidence suggests that high intake of carrot, Tiger lily buds,Chinese date, Honey and other fruits have been associated with lower risk of allergicskin diseases. Mustard seed (MS), namely Sinopis Alba Linn, belongs to the CruciferaeBrassica. Its elaborated products are favorite spices and seasonings which are widelyconsumed around the world. Recently mustard seed and its components have beendemonstrated to posses anti-inflammation activity. Allergy actually is immune specificinflammatory response. In Dermatology clinic, the most common is I hypersensitivityand â…£ hypersensitivity. However, there is few studies regarding the use of mustardseed to prevent allergy, and its anti-allergy mechanism is not clear. Based on the above background, the purpose of this study is to investigate thepreventive effects of mustard seed on type I hypersensitivity of skin and systems ofmice and type â…£ hypersensitivity, and to explore its anti-inflammatory mechanisms.This study will provide experimental evidences for mustard seed to be used in theprevention of allergic skin diseases in the future.Methods1To investigate the effects of mustard seed on type I hypersensitivity.â‘ a capillary permeability increasing model of mice using histamine andCompound48/80was established. Then the effect of mustard seed on capillarypermeability was detected using spectrophotometer colorimetry.â‘¡type I hypersensitivity model was established by intraperitoneal injection of OVA,then the IFN-γã€IL-4and IgE in the serum were detected by ELISA.2To investigate the effect of mustard seed on type â…£hypersensitivity.â‘ Model: an allergic contact dermatitis model of mice was induced by2,4-DNFB.â‘¡Experiment indexes:24h later, the swelling extent and weight of the ear weremeasured, then the rats were sacrificed to collect tissue samples from the ears andblood for histopathological and immunohistochemical staining, RT-PCR, andenzyme-linked immunosorbent assay.Results1The effects of mustard seed on restraining capillary permeability caused by histamineand Compound48/80The results indicate that mustard seed inhibits the increase of capillarypermeability caused by histamine and Compound48/80. There are significantdifferences between mustard seed group and control group (p<0.05).2The effects of mustard seed on type I hypersensitivity induced by OVAThe results of ELISA shows that the differences of IFN-γ, IL-4and IgE in theserum between model group and control group are remarkable (P<0.05), but thedifferences between model group and mustard seed group are not remarkable (p>0.05)3The effects of mustard seed on type â…£ hypersensitivity induced by2,4-DNFB In mice with ACD, mustard seeds significantly reduced the ear swelling,improved the tissue histopathological changes, suppressed the antigen-presenting ofLC, and reduced the mRNA expressions of IL-1β, TNF-α and IL-6in the ear(P<0.05)and TNF-α in the serum(p<0.05).Conclusions1Mustard seed inhibits the local cutaneous type I hypersensitivity of mice induced byhistamine and Compound48/80..2Mustard seed cannot affect the IFN-γ, IL-4and IgE in the serum of type Ihypersensitivity of mice induced by OVA3Mustard seed reduces the ear swelling of allergic contact dermatitis of mice inducedby2,4-DNFB.4Mustard seed inhibits the allergic contact dermatitis of mice induced by2,4-DNFB.The mechanism may be is related with the suppression of antigen-presenting of LC andthe reduction of the mRNA expressions of IL-1β, TNF-α and IL-6in the ear (P<0.05)and TNF-α in the serum (p<0.05). |
