OBJECTIVES: To investigate the inhibitory effects of Genistein(GEN) on mouse allergic contact dermatitis(ACD);To investigate the relationship between the inhibitory effects of GEN on mouse ACD and the balance of type 1 and type 2 cytokines through detecting the levels of type 1 or type 2 cytokines produced by draining lymph node cells and offering experimental basis for the clinical application of GEN on the disease caused by type IV allergic reaction such as ACD. METHODS: The animal model of ACD was induced by DNFB, the effects of different doses of GEN(8, 20, 80mg/kg) on mouse ear swelling, histopathological changes of mouse ear skin, thymus index , spleen index , body weight and the toxic reaction were observed .The levels of type 1 cytokines (IFN-γ, IL-2, TNF-α) and type 2 cytokines(IL-4, IL-10) in supernatant of cultured draining lymph node lymphocytes in mice were measured with LuminexTM 100.RESULTS: All groups of GEN inhibited mouse ear swelling induced by DNFB significantly, reduced the edema of epidermis or dermis, inhibited the infiltration of inflammatory cells, decreased thymus index (P <0.05), the middle or low dose of GEN increased spleen index (P <0.05), high dose of GEN decreased spleen index(P <0.05); GEN did not affect the increase of mouse body weight(P <0.05), had no obvious toxic reaction. GEN inhibited the up-regulating of IFN-γ, IL-2, TNF-α , IL-4, IL-10 in supernatant of cultured draining lymph node lymphocytes in sensitized mice significantly(P <0.05).CONCLUSIONS: GEN can inhibit mouse ACD induced by DNFB significantly; GEN affect the relative weight of thymus and spleen significantly. GEN can inhibit the up-regulating of type 1 cytokines (IFN-γ, IL-2, TNF- α ) and type 2 cytokines (IL-4, IL-10) in supernatant of cultured draining lymph node lymphocytes in sensitized mice significantly, which might be one of the mechanisms that GEN inhibit mouse ACD.
