Purpose:Observed with stress-induced depression of Yanggan Jieyu particles of chronic liver injury in animal models of serum liver function (GGT, ALT) and content of plasma neuroendocrine (ACTH, BDNF), the content of the hippocampus5-HT2AR positive neurons expressingexplore the mechanisms of the pathogenesis of chronic hepatitis with depression and Liver stagnation particle treatment of chronic hepatitis with depression.Methods:1.60mice were randomly divided into control group, fluoxetine hydrochloride capsule group, low particle of Yanggan Jieyu in the high dose group, a total of five groups (n=12);170rats were randomly divided into control group, liver injury model group, liver injury with stress depression model group, Yigan Yiqi jieyu particle group, fluoxetine hydrochloride group and Liver stagnation particles, high dose group, a total of eight groups, fluoxetine hydrochloride capsules group30and20of the other groups.2.line I mice tail suspension test; solution subcutaneous injection of40%to rat CCL4, oil (1ml/kg,2times/week for12weeks), the composite made of the combined singly housed and chronic unpredictable mild stimulation model approach (per cage a day randomized to reverse day and night, the ice water swimming, fasting, water deprivation, electric shock, heat drying, the tail clamp to stimulate a continuous21days), the establishment of chronic liver injury with stress in the rat model of depression.3.was measured in the tail suspension immobility time;, serum liver function (GGT, ALT) content of plasma neuroendocrine (ACTH, BDNF), content changes and brain hippocampal5-HT2AR positive neurons The expression levels.Results:Yanggan Jieyu particles can significantly shorten the tail suspension immobility time of treatment with fluoxetine hydrochloride;Yanggan Jieyu particles to improve liver function, regulate the content of neuroendocrine reduce the hippocampus of experimental rats-HT2AR positive neurons in the role of the average gray value,the efficacy of high dose group,and the overall effect is better than to Yigan Yiqi Jieyu particle group and fluoxetine hydrochloride capsule group.Conclusion:1.The composite modeling method can be successfully prepared by chronic liver injury with stress depression in animal models.2.Yanggan Jieyu particles to treat chronic hepatitis with depression and its hepatoprotective reducing enzyme, the mechanism of antidepressant may improve liver function, regulating neuroendocrine content, lower positive neurons in the hippocampus5-HT2AR average gray value.3.Plasma ACTH content increased, the reduction of BDNF content may be the pathogenesis of chronic hepatitis with depression.
