| Objective Learn by observing the pain behavior changes of the chronic CIPN rat model,observing the expression changes of NR2B in spinal cotd dorsal horn and pNF-H in dorsal root ganglion neurons,to explore Dangguisini soup on the preventive efiect of oxaliplatin induced chronic CIPN.Methods52adult Wistar female rats were randomly divided into five groups: blank group,model group,control group,low and high dose group of Dangguisini soup.Waiting for the preventive administration of seven days,in addition to the blallk group(n=8)injected intraperitoneally to5％GS at the dosage of5ml/kg body weight fron d8to d30,the other four groups(n=11)injected with oxaliplatin at the dosage of4mg/kg body weight twice a week.Blank group,model group and control group were receiVed1ml of0.9％NS by gastric Iavage daily from d8to d50；and Dangguisini soup group were received high and low doses of the Dangguisini soup at the dosage of5ml/kg body weight by gastric lavage daily；control group were received Methylcobalamin at the dosage of104ug/kg body weight twice a week.The rats werc weighed every3days,observed the changes of reactions,diet and stool daily, measured MWT and the latency of the tail thennal hyperalgesia once a week.Waiting for the The fiftieth day,each group of rats were dissectioned,and removed the spinal cord,L5dorsal root ganglion. the expression changes of NR2B protein in the spinal cord was detected by RT-PCR and Western-blot to observe the preventive effect of Dangguisini soup；and the exptession changes of NF-H protein in DRG was detected by immunohistochemical to express the impact of the soup to oxaliplatin induced CIPN.Results The success of that chronic CIPN rat model prcparation was proved by the bellavior measure and histological examination.It was showed the TCM group have significant difference(P<0.05)compared with model group in weight and behavioral tests, but the difference between treatment group was not significant(P>0.05).the expression changes of NR2B protein and mRNA in the TCM group have signincant difference(P＜0.01)compared with model group,have no significant differences(P>0.05) compared with blank group.The immunohistochemical show the expression of pNF-H in the TCM group was increased compared with the model group,was similar to blank group.Conclusions the expression of NR2B protein in the TCM group was decreased, the expression of pNF-H was increased, and have clear correlation with behavioral manifestations, suggesting Dangguisini soup plays an important role in the prevention and treatment of chronic CIPN. |
