ObjectiveFirst, we reviewed the general situation of studying hemorrhagic stroke with traditional Chinese medicine,then we analyze the theoretical basis of Didang Decoction treating hemorrhagic stroke. Second,we research the influence of Didang Decoction on hypertensive cerebral hemorrhage model rats’behavioral neuroscience, regional cerebral blood flow, the expression of brain tissue hypoxia-inducible factor-1α, transferring/transferrin receptor,and we discuss the mechanism of Didang Decoction treating hemorrhagic stroke.MethodThe study include theoretical part and experimental part.Theoretical part:We collect the literature of hemorrhagic stroke with traditional Chinese medicine and the study of hemorrhagic stroke with contemporary Chinese medicine systematically,then we clarify the theoretical basis of Didang Decoction treating hemorrhagic stroke.Experimental part:We Reproduced hypertension model by intraperitoneal injecting posterior pituitary,and intracerebral inject autoblood to establish cerebral hemorrhage model.We interferential treat hypertensive cerebral hemorrhage model rats by using low-dose Didang Decoction,middle-dose Didang Decoction,high-dose Didang Decoction respectively seven days. Then we research each group’s alteration of neurological score,regional cerebral blood flow,and apply the method of immune histochemical,western blott, polymerase chain reaction to detect rats brain tissue HIF-1α, Tf/TfR.ResultsTheory research:Activities of Qi ascent-dascent disorder is the key point pathogenesis for hemorrhagic stroke.Brain veins injury is the basis of pathobiology for hemorrhagic stroke.The blood to be detained in Intracalvarium is the centre of pathobiology for hemorrhagic stroke. Activating blood circulation to dissipate blood stasis and purgation is the important method of treating hemorrhagic stroke.Experiment research:①Neurological score:the scores of Didang Decoction treatment groups are notablely lower than model group on the3rd day,the7th day (P<0.05).②rCBF:the rCBFs descent range of Didang Decoction treatment groups,especially Didang Decoction low-dose group are notablely less than than model group on72hours (P<0.05).③HIF-1α detection:the HIF-1α positive cells, HIF-1α protein content,HIF-1α mRNA expression of Didang Decoction treatment groups,especially Didang Decoction low-dose group are notablely higher than model group(P<0.05).④Tf/TfR detection:the Tf positive cells, Tf protein content,Tf mRNA expression of Didang Decoction treatment groups,especially Didang Decoction low-dose group are notablely higher than model group(P<0.05).The TfR positive cells, Tf-R protein content,TfR mRNA expression of Didang Decoction treatment groups,especially Didang Decoction low-dose group are notablely higher than model group(P<0.05).ConclusionThere are theoretical basis, experimental basis and clinical basis to use Didang Decoction treating hemorrhagic stroke. So we think that Didang Decoction is the effective prescription to hypertensive cerebral hemorrhage.Didang Decoction can reduce neurological score of hypertensive cerebral hemorrhage model rats, and raise rCBF of hypertensive cerebral hemorrhage. Didang Decoction can raise the expression of HIF-1α,in order to increase rCBF of hypertensive cerebral hemorrhage to improve the ischemia and anoxia condition of brain tissue. Didang Decoction can raise the expression of Tf/TfR, in order to speed up cleaning Fe2+, for confronting neurotoxicity and reducing encephaledema. Didang Decoction can up regulate the expression of HIF-1α, Tf/TfR, in order to protect neurons,for encouraging nerve plerosis of hypertensive cerebral hemorrhage model rats.
