The Mechanisms Of Chinese Medicinals With Functions Of Tonifying Kidney And Nourishing Yin For Treating Perimenopausal Syndrome Study

The symptoms occurring in women before and after menopause, around menstrual disorder or menopause, including dizziness, tinnitus, baking fever, sweating, restlessness, liable to be angry, tidal fever, red face, palpitation, insomnia, or soreness in the back and lower back, swollen face and limbs, anorexia, loose stool, or ant-crawling sensation on the skin as well as unstable emotion and mind, are called various kinds of symptoms before and after menopause, which pertains to the category of modern medicine“perimenopausal syndrome”. Modern medicine believes that perimenopausal syndrome refers to a group of symptoms characterized by functional disorders of vegetative nervous system due to declined ovarian function and decreased estrogen level. The medicinals for tonifying kidney are frequently selected in traditional Chinese medicine (TCM), whereas hormone replacement therapy (HRT) is mainly adopted in modern medicine. Although HRT has confirmative therapeutic effects in alleviating clinical symptoms, its application is restricted because it can result in some adverse effects like estrogen-dependent malignant tumor, impaired heart and stroke. Chinese medicinals has some advantages for treating perimenopausal syndrome. It is not only effective, but also safe. From the aspects of TCM theory, modern medical research and pharmacological experiments, the mechanisms of Chinese medicinals with functions of tonifying kidney and nourishing yin for treating perimenopausal syndrome are comprehensively illustrated in this paper.Combined literature and clinical research, this paper holds that insufficiency of kidney essence and essence failing to transform into blood are crucial etiology; kidney deficiency as the root and yin-essence depletion are specific pathogenesis; the kidney is the major diseased position, and other zang-organs such as liver, heart and spleen can also be involved as the diseased course prolongs. Therefore, Chinese medicinals with functions of tonifying kidney and nourishing yin should be applied to treat this disease. The key therapeutic principles are tonifying kidney and replenishing essence, then regulating liver and nourishing blood, and other zang-organs should also be combined.In order to reveal the action mechanisms of Chinese medicinal preparation Menoprogen Capsule with functions of tonifying kidney and nourishing yin, the study adopts modern experiments, drug efficacy and pharmacological methods, Ovariectomized rat model (OVX) was established and estradiol (17β-estradiol, E2) was used as positive control.The action of Menoprogen Capsule was observed.Results:Both Menoprogen Capsule and E2could significantly elevate the uterine coefficient of OVX rats, increase the level of serum E2, indicated that Menoprogen Capsule has estrogen-like effect. Menoprogen Capsule can up-regulate the expressions of VEGF, AQP2as well as AQP5in uterus by immunohistochemical method. E2, Not as E2Menoprogen Capsule couldn’t promote the expressions of ERα, PR, c-jun and c-fos in OVX rat uterus.And yeast two-hybrid assay showed no components combining with ERα or ERβ in Menoprogen Capsule. Menoprogen Capsule can be used to treat menopause syndrome. The risk of estrogen-mediated endometrial cancer is low for this treatment because its mechanism is different from estrogen-like substances,which provide scientific evidence for clinical treatment for menopausal syndrome.
