Sjogren’s Syndrome (SS) is due to abnormal cellular and humoral immune reaction, causing lymphocytes and plasma cells infiltration around Glands and tubules, hyperplasia of connective tissue, inflammation and destructive lesions of the patients. The symptom includes dry eyes and mouth, multi-organ damage. The etiology is still unknown. The modern medical treatment of the disease mainly focuses on replacement therapy, which is unable to cure the disease fundamentally.Traditional Chinese Medicine shows its potential and advantage in the treatment of SS. During the clinical practice of the research group, according to the theory of "dryness pathogen injuring yin""body fluid and blood sharing the same origin ""Blood being the mother of Qi", and with the fact that about half of SS patients displaying symptom of salivary gland inflammation and swelling, also combined with the experience of Chinese medicine treatment of surgical sore, the research group develop Ganzao liquid (referred to as GZY) with overall clinical efficiency of96.7%. In order to investigate the mechanism of GZY, we conducted the following experimental research.Objective:To develop SS mouse model by immune induction, feed them with Ganzao liquid, which supplements Qi, nourishes Yin and reduces phlegm. After observing the expression of ICAM-1, serum TNF-a level, then verify Ganzao liquid can have immunomodulatory effect, which is the effective TCM prescription for treating SS.Methods:After developed to SS model, these mice were divided into5groups (normal group, model group, Dandiqiongyu group, GZY-low dose group, and GZY-high dose group). Observe every mouse in fur,weight, food-intake, the amount of drinking water, immune organ index, the amount of saliva and the pathological changes of Submandibular gland; use ELISA method to detect concentration of serum TNF-a: And use Immunohistochemical Method to detect expression of ICAM-1of Submandibular gland Results:1. Compared with normal group, the fur of SS model mouse is fluffy and loose, with gloss decreased and irritability; the food intake of SS model mouse is almost equal to normal group mouse’s, but the weight growth trend is obviously lower than the normal group mouse’s; the water intake of SS model mouse is more than the normal group mouse’s. After observing the saliva flow, we find the saliva flow of SS model mouse is obviously lower than the normal group mouse’s from the third week and till the end of the experiment, speculating that the SS model mouse feel thirsty.2. The submandibular gland HE staining of the SS model mouse displays the symptom of interstitial, lymphocytic infiltration around duct and vascular, lymphocytic foci formation, catheter dilation and atrophy, wall hyperplasia lesions.3.The immune organ index of the SS model group is larger than normal group’s. The organ index of the treatment group is lower than the model group, the spleen index of high-dose GZY group is significantly lower than model group.4. The normal group has lowest concentration of TNF-α, and the model group has highest concentration of TNF-α in all groups. The high-dose Ganzao liquid group has significantly lower concentration of TNF-α than SS model mouse, indicating that GZ,Y can reduce the level of inflammatory factor.5. Normal group mouse has no or little ICAM-1expression on submandibular acinar membrane and Duct endothelium. SS model mouse has significant ICAM-1expression on these locations, which is in accordance with the severity of disease. The expression of ICAM-1of GZY group is between normal group and model group, indicating that the GZY can improve the SS’syndrome partially.Conclusion:The GZY may improve SS’symptoms by suppressing the enhanced immune response, reducing lymphocyte infiltration in the salivary glands, alleviating the invasion of inflammatory factors, decreasing the level of ICAM-1of salivary glands and TNF-α of serum.
